A more complete update of this bunkology. 1 • 28 • 20• World Map - Day 29 • In Sequence:Spore Syringe → Germination Plate → 5x T1's → 9x T2's from 4x T1's / 1x A2G from 1x T1 • T2 Day 14 & 10 •
 (top) T1 Plate 3 and its T2's. Seems the selective transferring did in fact subsequently represent the 'desired' growth. (bot. left) T1 Plate 1 and it's T2's. (bot. right) T1 Plate 5 and it's T2's. Maintaining the same patterns. Things that I've thought may have been foreign contaminants, I now wonder if were bits of the myc colony trying to run only to be tethered back in by something. As my frustrations with this culture pool with each passing day, I'm considering a 'hail mary' - make a Liquid Inoculant from one or more of these T2's and inoculate a pint of rye with them.• A2G Day 9 • For 9 days in, this is moving like frozen molasses. But it is moving. Will be interested to see what comes of a shake at 20% colonization. There's almost certainly bacterial intervention - made evident by the uncolonized, wet grain pressed against the glass and the slow growth. Also suspect to be the cause of the odd growth characteristic.
1 • 27 • 20• Germination Day 3 • Only one plate shows any spore germination which it, again, showed it the first 12 hours. I fear it's displaying the same characteristics as the first trial. Seems the swab streaking was effective - you can very obviously see the trail of bacterial bits deposited away from the desired culture. Quite the frustrating culturing effort. Maybe the spores are old and tired. Maybe there's a suppressive bacterial culture in there. Maybe I'm in the wrong jungle...

I just bought a case of 1/2 pint smooth side jars and I know whatcha mean  I got them to make PF cakes to test LC's on, but now I wanna fill them with wbs and all kinda shit using one for LC now

How will you expand? 1:3 1:2?

I've got a bunch of half pints with SHIPs from my first PF Tek grow, and I often use them much like this when I have extra grain. The only difference is that I like to use them for grain liquid inoculates down the road. (The super-simplified tek: let grain colonize, shake, inject 60ml or so of sterile water, shake again, aspirate LI, knock up a bunch of jars or bags.)
If I'm knocking up a test jar of a new culture, for example, I may also inoculate a half pint like this, then toss it in the fridge once colonized while I grow out the test jar. That way, if the test jar turns out well, I have the option to immediately knock up a bunch more without having to grow out more agar plates. And colonization is also generally super quick from GLI - ime, 5-7 days is typical.

I haven't g2g with 1/2 pints before but I'd probably go at least 1:4 maybe even 1:6 if I'm going through the trouble. G2G goes a long way.
Durgin that sounds like a cool idea probably much faster for expansion. Historically I don't do well with liquids so I don't use them really anymore.

I got the idea from Ziran's tek, https://www.shroomery.org/forums/showflat.php/Number/24769612. Only difference is using the SHIP instead of opening jars. Unlike LC, you don't wait before using it, so even if something gets in there isn't time for it to expand before the myc starts colonizing. It's not foolproof, but assuming you can successfully sterilize a big syringe of water, there's no more contamination risk than doing g2g (and probably less, since there's less exposure to air during inoculation, whether through ship or squirting under a cracked lid).

Me too. My master quarts are 1 to 10. Honestly IDK WTF I'll do with these. I spent 5 hours last night and all morning today doing myco work I'm spent for a while. Should probably go to the gym or something lol.

First of all, I just have to tell you guys how awesome you are and how much I appreciate all of you!
I've spent some time reading the agar threads and getting familiar with what to look for on my petri. I want to get really good at isolating my species. This is my first time using agar and I found i to be pretty easy (bye-bye syringes!). I inoculated my petris with a spore syringe and these are my first transfers. I'd like to do another transfer and need your experienced eye as to what to take.

The spots at 4:30-5 were caused from droplets of condensation water after I set a cup of hot water on the dish to remove the condensation. I don't think it's bacteria because they were not there before I put the hot water on the dish. But I'll defer to you!

Me too man. I have full time job, wife, 8 kids, 2 dogs, 2 cats, squirrel, hampster. My life is so packed full of "life" I don't have mush time to journal. I tried to catch up. I've shoke the Tailfeathers and Golden Halo. Noced up 3 more jars each and 3 RW. I also have LPEU, ape clones, pe6 clones, Costa Rica clone, melmak, and AA+. I'm slowly transitioning to bags. I've got alot of jars. I mixed up my melmak bags. They was like 50% already so fingers crossed they recover good.

Few of my plates. Loving melmak, got the pesa and toc mostly tamed with one rhizo one tomentose isolated on the toc, but man this aa+ is wild.

If you're talking the stuff in between the two transfer pieces, no, that looks like mold. I would transfer some of the rhizomorphic mycelium around 6 o'clock on that piece ASAP. Those spots do look like bacteria to me. I'd go ahead with a transfer from both of them while you're at it and then let that transfer grow out some to see if it sectors further, as long as other contams don't pop up.

All my agar growth looks weird. The germ plates all had Mycelium traveling under the surface. I took transfers of the best parts but the t1 plates all look really weird too.
I'm gonna see if I can get it growing proper but I may have to start over. Golden Halo and RW plates look like this, maybe I made that agar batch funky.

Also I'm getting nothing from my tamp print...

Well that's a total bummer and it looks like you've been around a long time. Can you post the agar recipie, brand of agar? Well I see in your log lots of choices for agar...

I teaspoon agar powder/100ml water. The germ plates were without nutes, just the sugars they put in this box agar. The t1 plates are same recipe but using grain water.

Last night I made up some new plates using the same portions but using 25ml beer/75 water. I threw in some rice while boiling it and I could see it leached out into the agar. I poured plates minus the rice chunks.

I've had this project on the back burner bc I've got a lot of others to tend to but now I'm focusing so I know I'll pull it off. My only guess is that I prepped up a bunch of pitris and they sat around a few weeks. Normally that wouldn't concern me but I'm using a new (to me) presto stove top pc. I only ran a 20 min cycle on those pitris and maybe something lived. I did see bacteria in one after the t1 xfer. It has been working really well for me until this. Could be bc I smoke and drink and make mush all night too...
 I still have clean original germ plates to work from so tonight I'll start fresh plates and I'll see what I can get these t1's to do.
