Cracking grain jar lid to inoculate MS

I've seen that people open the jar to drop in a wedge of agar in-front of a flow-hood or inside a SAB.
I'm assuming I can also use this method for injecting via MS syringe?
(yes I know not preferred and I do currently have some agar plates going on its 1st transfer and I'm well aware agar is the way to go before anyone bashes MS to grain)
However until these agar plates and future agar plates are ready I was going to innoc some jars up with MS.
I figured cracking the jar lid eliminates the need for SHIPs and also the needle doesn't actually touch anything so I can just spray some solution around the grains on top.
Makes lids easier to make as I can use one piece lids with a hole in the top and just a full SFD underneath. No silicone no glue no nothing.

I’d just stick to poking the syringe through the GE hole, then mp tape over it. That’s how I did syringe to grain for a while it’s really easy and way safer. It only takes a second to glob some silicone ontop too for the ship.

I’d only use a couple of drops per jar. It might cut back on contamination.

Innoced some jars last night.
Plastic lids 1/4 inch hole in middle melted with soldering iron.
Full SFD underneath.
PC for 90 mins with lids a little loose.
Remove from PC and tighten lid but not really tight.
Put in SAB to cool down.
Loosened lids then flamed the needle and cracked lid and squirted in.
A lot easier than SHIP.
I also transferred my first PE agar plate to 3 others so moving forward will be doing afar wedge to grain with the same lids

Update on the PE agar plates (1st time)
I had one plate that looked good and transferred a small triangle to 3 plates.
This was the original...

These are the 3.
One...

Two...

Three...

Which plates would you transfer from and where and to how many new plates?
Thanks,

Thanks, I've only got 3 plates left at the moment so I'll take 3 transfers from the first two plates at 12 o'clock onto 3 new plates.
Would you normally do another transfer after this one or does it depends on how it looks?
As these are my first plates and new to this I have a question that could be stupid... can I just continually keep transferring from plate to plate or does the myc "get old" or something after time?
Thanks,

Yeah you can transfer for a long time before it gets weak. Check out verums hunting PE clone thread. I think he went over 100 transfers trying to make monocultures.
It’ll depend how the next plate goes. Most of my agar has been with clones, my first spore plates were LAGM. With clones I would have a good plate by t2 probably half the time and almost always by t3.
With ms it’s taking a little longer. I’ve had a bunch of solid t3s but a few are gonna need 4 or 5.
Once it’s got solid uniform growth it’s good. I’ve had it take 5 plates and I’ve also had it direct from clone tissue.
