First attempt at agar when should I make a transfer????w/pics

On 01/20/20 I used a Ms syringe of PE & Ms syringe of GT on 2 pre made malt extract agar plates each. This is my first go at Agar was kinda just messing around with it since I now have a nice SAB!! The reason there is a couple spots of mycelium growth on a couple plates cuz my dumbass didn’t wait long enough for the needle to cool when squirting to agar and it kinda of made it squirt/ mist all over lol. I thought the heat might have killed everything at first for like 5 days I saw no signs of anything at all. Even then it was only on 1 plate of each, but now all are showing the last two especially the one GT is just starting so I can’t even get a good pic yet. Pics are hard to get what I’m seeing by eye but The growth on a couple even seem to be rhizo. So far I can’t see any signs of contam but my eye isn’t trained to see what a lot of you OG’s see! I just want to know how big should I let it get until making a transfer, any advice or tips for me? I know it’s probably going to be hard for anyone to even say much because the pics are so hard to get good quality thru the plate and the plastic bag.

I would make the transfer to a new dish when the culture is about the size of a dime. The reason being is since you inoculated with spores contaminates are not too far behind.I would also recommend learning how to pour agar since pre made petri-dishes are expensive.

Ok cool thanks. Yea I actually just ordered everything to make my own dishes from now and as well as got myself a bunch of Wbs and different grains to try as well as a couple pressure canners so I can start making my own jars, etc. from now on. I just started getting into mycology, so I did buy everything premade, jars, bags, plates, etc! Now that I’ve got my feet wet, everything’s going good and I have 3 jars and 10 bags a couple weeks into colonization. I now feel as if I have done enough research to start diving in head first. I feel a lot more comfortable making everything myself for all future projects now (Which is a Hell of A lot cheaper for sure).

I am going to do my first transfer plates today on atleast 3-4 plates I did. Out of the 4 plates this last one which took the longest to show and now just looks the most different from the other ones. Need to know if this is good mycelium or something else at this point. I’m still trying to get the eye for contams down as I’m still very new to this!

Unless you sprinkled the spores on like this you shouldn't be getting anything growing on the perimeter. That is where 95% of agar contams are at. Probably sucked in cooling.

No I fucked up and when trying to get the slightest amount possible squirted out of the syringe while it was too hot resulted in spores not going directly center as intended and basically they misted the plates/squirted everywhere! That’s why I’m not sure with these

Looks like a good chance those satellites are myc.
All the mold I get has turned colors before getting as big as the one on the gt pic.
Smaller ones could be anything but you’ll know in a day or two.

Not all molds will be that obvious on agar. I wouldn't trust that growth on the edge. Maybe if it looked really really good but i don't ever put anything around the perimeter so i don't have to guess like that.

Yea def a lesson learned. Will def make sure while doing transfers today, to get it right in the center this time! I’m pretty sure it’s mycelium but I’m still a Noob and idk going to wait a day or two and see how it looks before I decide. I have another GT plate which is much better anyway. I was just interested in everyone else’s input. Thanks for the responses tho guys, much appreciated.

The ol hidden plate trick. I see how it is. Well, there ain't no harm in making a transfer and seeing what happens. Or let it roll and see what happens. The main thing is just to grow shit and pay attention to what does what, why and how. You'll be a master in no time.
