How many sectors should I transfer away from this Mazapatec clone on agar?

Hi there. This is a tissue clone put onto agar 40 days ago. It seems to have various sectors and I was initially planning to take transfers from all 5 zones marked in red pen near the perimeter of the dish. Although the mycelium is thin and difficult to see, it does reach out there. Running short of petris and time, I just did a single transfer from 3 o'clock (the right-hand side). Mother and baby both doing fine :smile:



I am new to this hobby, so I am wondering if it is worth taking additional transfers from one or more of the other sectors. Do you think they would grow differently?

I don't really need any more clone strains right now, as I have been doing MS BRF cakes galore, so I have lots. I am just curious whether the various sectors on this petri might behave differently, or if they are basically all the same creature. I am just trying to learn. This is a tough hobby!

Yes, it was put onto agar 18th December (I dated the dish). Not sure why it's growing slowly, but I think my agar might have been lacking some nutrition. I am experimenting with various recipes. Potato boil water working better for me now! Also am adding food coloring to make it easier to see now.

All the growth on the agar plates in that batch was very weak and hard to see. It looked to be quite well organised though, when you got it in the right light.

my go to is MEA water  8:10:500- 10:10:500 is slower - 6:10:500 quicker

Thanks for the recipes. My last batch of MEA plates all contaminated with bacteria growing all over the place. I know my LME was out of date so that might have been the cause. I will try it again with different packet of LME.

6:10:500 quicker? I guess that is LME:agar:water?

So they grow more slowly with more nutes - that makes a lot of sense! Maybe I added too much nutes to the last agar and that is why they grew slowly. Must check my notes - maybe I wrote it down, maybe not.

I switched to an antibiotic agar mix and would never look at doing it differently. I’m not sure if I can post the name of the product source. But if you look at the world of Mushrooms and psychedelics his name is at the top of the list as a speaker. Wink wink...

Its doesnt matter if something is out of date.  You could 100% sterilise a dead rat and it wont contaminate unless a mold spore or bacteria gets in there.

All recipes work but its whats quickest and clearest for me.  These plates are 8:10:500

Yeah 40 days... That timeline and the bacteria around 5:00, I'd say it's bacterial. I'd use black tea agar for the next round of transfers.
