Best Liquid Inoculant (LI) for Inoculating grains FAST. 1

What are the best procedures to make a wicked LI syringe to knock grain fast?
I'd like to narrow this to how to make it in a syringe (I pre-seal bags and own no flowhood)
I am currently:
1) Pouring thick/deep plates so the LI is thicker and soupier. 2) Adding a bit of Karo to my pre-purchased Malt-Agar, hoping sugar assists leapoff. 3) Making my agar soft. 4) Blending it as little as possible, but enough that I can suck most of it up though my basting needle (seen in photo). 5) One plate, 50cc of water, one huge (50cc) syringe, injecting two bags (gallon each)
Curious about:
a) Letting the LI sit before use b) If I am over-blending it. (most of the pieces are blended to under 2mm) c) If its possible to over-mix the LI and grains, damaging the mycelia even more.

I think brf slurry is fastest I've seen but would be excluded by pre sealed I guess. Don't think any of your concerns are something to worry bout.
Using a 2 mm wedge? Try using a huge piece. 3/4 or a whole plate if your confident it's clean. Make the agar super soft and you won't need to blend it. You'll be able to shake it apart.

I'm currently gettin colonization around 14 days on average. I want it faster than that. I'm just trying to get it the best it can be.

I'm using a whole plate. Blending to 2mm.

I'm well versed in making LC. I just want to have this method down when I got a plate or two that I want to use that day.
Does anyone have thoughts on the "letting it sit" and maybe coagulate a bit?

I have used this tek more than 10 times successfully. Like you I don’t have a flow hood, I’ve had no issues drawing up 10cc poking through bag, taping pone hole with two layers Micropore, my bags are fully colonized no more than 8 days, sometimes sooner.

I usually blend up a plate in a blender jar then suck it up into syringes. Direct culture injection. One 1/4 pint water to a plate will make alot of li. I beleive i used to make 6 culture syringes from 1 plate. And then you can put one drop on a few plates to get the culture expanded before you inject the whole syringe into bags or grain jars. Ive also stored cultures like this for 5 plus years with good performance. After that it slows down dramatically.
The thicker the better. When its thicker it sticks to every piece of grain creating more inoculation points.
To put it in layman terms this would be my notes:
1) slurry 3-4 days.( account for 3-5 days to colonize brf jar with clone syringe.) 2) Cornmeal slurry 4-5 days. plate has to colonize 3-5 days 3) LI blended from a pasty plate recipe. 7-10 days. 14 on the slow side from a bad shake or pour session. Also account for 3-5 days for a plate to colonize. 4) LI blended from standard agar on thin plate. 10-14 daysz also account for 3-5 days for agar to colonize plate. 5) i havent done lcs in so long i couldnt tell you. But you have to transfer culture then let lc colonize. So add in those factors. The reason slurries work so well is the thickness. Lately ive noticed that pasty recipe with more agar seems to colonize faster than a standard agar mix on a normal thin plate.
Factors to conisider. Wbs colonizes much faster than oats or rye. There are far more inoculation points.

These took 4 days to colonize. I shook the two on the left, the right one was never shaken. Pint Brf cake>slurry in a hurry tek. Those are gallon jars.

The surest way to gain speed and performance imo is to focus on cloning and culture work. Have seen clone grows on this site that went from plant to harvest in only 17 days. 12-14 days with agar LI seems pretty standard, the only factor I've personally seen make MS slurry jars faster was increasing the temperature in the room they were stored in to about 73-75f, which shaved off a day or two.

Boy I'm feelin that. I just started anew with spores because of a piggybacking contam and my MS mycelia is slow.
But while that is true, I am looking at my perfectly prepped winter wheat berries in a bag and can't help but think that I can get an agar plate to leap faster than I am currently doing. I wonder if a bit of coffee added to my berries pre-PC'ing could coat the husks with some goodness...

Call it paranoia/preventative maintenance. Jars come out of the PC and go in the shmuvbox , I work and then seal them in the shmuvbox . When I get ready to use them I peel off the press-n-seal and alcohol wipe around the lid, in the shmuvbox . The lid and seal never gets touched by hands or open air until I break the seal under flow. I handle my jars a lot and I just don't want anything to be able to get in that gap around the lid. Not necessary but I'm having consistent success and doing this isn't going to hurt anything so I'mma keep doing it.
 ^^These are AA+ started Saturday from one slurried pint jar. Multispore. There's 4 more gallons as well. Started the gallon jar on Friday. My 'jane doe' project, multispore from a pint pf cake. There's six more gallons of 'jane doe' as well.
 ↑↑The pint with the ? is the one I slurried to 4 gallons plus what you see in the first pic. I just forgot to write the date hence the '?'.

I was thinking.... what if I left one of my LI syringes at room temp for 3-5 days, then nocced my grains.
pros: my agar might get more gelatinous and stick to the grains more.
pros: The mycelia might grow (sorta like LC does in the broth)
cons: Could get contamed and couldn't see it, and sorta defeats the purpose of LI.
