MS to agar - noob question

Its my second attempt at agar. And it is the second time i see no growth or contamination on my plated. I have no idea why. Ive followed the c10 agar TEK.
So my question would be, does my tap water HP levels have anything to do with it. Secondly, how long does it usually take before you see and growth or contamination.
And thirdly, where do you store your agar (what temp.)?

How did you inoculate the plates?
Could try distilled or spring water your PH could be the issue.
Spores can take upto 3 weeks to germinate, contamination has to be introduced it won't just appear.. Bad sterile practise could do that
Anywhere from 18-28c will be fine temp, higher end being preferred for a speedy growth
Can store them anywhere once there sealed, I stack em on a shelf out of direct light

I've inoculated directly from the MS solution into a syringe and onto the agar. For the agar i used (per. 500ml: 10g agar-agar, 10g potato dextrose and 3.5g dextrose)...and some food coloring. Its been 68h since the inoculation, Im keeping them at 20C in the dark.
How much do you think the tap water is a factor? Should i bother testing the PH level, or just switch to distilled?

If it has only been 68 hours, you are just not being patient. Give it some more time. Spore can germinate in a day or 2 or a week or two depending on the age of the spore etc. I would say if you don't see any growth in 2 weeks, start over.
I have in all the years not once ph balanced my agar and I haven't had any issues with germination. I don't use straight tap water, I use filtered water, bottled water or distilled water
I keep the room where all my agar plates are at 25C and that seems to be the best in my experience. That is what works for me

I agree - you just need to wait longer. I inoc'd my first 3 plates on the 15th, and 3 more on the 16th and only have growth on 1 out of six. I'm pleased.

Seconded, toss all the plates except for the first one. Take one sample from each of those larger colonies, you could get 6 or 7 total if you want.
