Much love all! First time agar 6 days after inoculation, any thoughts?
    #26439588 - 01/18/20 03:38 AM (4 years, 30 days ago)

:ooo: Hello everyone! 6 days ago i went the agar way for my first time :mushroom2:.
Tried to follow your advice and went SAB- agar went PC for 20 mins and poured them too fast so i had to remove condensation.Used a GT spore syringe that i wanted to save (failed with it 2 times inoculated grains straight) Everything went in the frigde and 1 day later introduced to room temp (my bad, lack of knowledge).
8 plates remained uninoculated in their sleeve unwrapped and have no sign of life.
9 plates seem to be colonizing most of them where the spored from the syringe landed (it was the end of the syringe so i could see the print coming out)
The rest have no sign of mycelium / bacteria either. So... should i worry about buying a blade ? ^_^
Also yesterday i did my second agar, poured 20 dished and inoculated 12 of them with 3 different strains. The agar seemed very dense (i have the strips agar) do you think is going to be a problem?
I'll try to post pictures, they are not the best but this is what i can do now. Don't forget: YOU ARE AWESOME  !

Lookin good!  It'll be time to do some transfers pretty soon.

When you say you had to remove condensation, how did you go about that?

In case you haven't seen it before, this is the tek I use to pour condensation free plates https://www.shroomery.org/forums/showflat.php/Number/21922023

So i tried to follow exact tek, used another bottle tho and to be honest my temp in the water bath did not go over 48C even after 10 minutes so i poured thinking it's ok. All got condensation, removed it with cup hot water but after everything went in the fridge so made condensation worse.
Yesterday was my second time and I waited longer for agar to cool and tried to pour 3 sleeves of 10 stacks small petri and by the 20th plate it  was solid. Even like this condensation still was there. Thank you for your reply!
I am trying to figure it out if i should toss any of the plates. They are all wrapped in saran wrap and individual ziplock bags.

The only thing to do with condensation is maybe pour in a warmer room, but really just stack the fresh plates as high as possible. The top two or three will always have pretty bad condensation but after that all clear.

Get a torch when you get a blade also, and make sure you get the whole blade red hot I was having a lot of bacteria cuz I would just get the last third of the blade glowing thinking that would be enough.

Cool!!!  :takingnotes:
About the second agar i realized some of the water evaporated when diluting the agar in water so that is why it is so stiff, would that be a problem ? it would be something like 10g agar 7.5 lmea and 350 ml water (where it is supposed to be 500 ml)

"You would ideally want to do a transfer from a dish as soon as you see growth to avoid spreading any contaminants during the transfers. The less there is growing on your dish before the transfer the better.
"
How many days should i wait before transfering any of them?

I would transfer now, from the leading edge, as far away from suspicious growth as possible. I'm new to agar also.

The 10 plates that i have left with agar for the transfers have really bad condensation, it is going to be a problem if i try to transfer to them? and if I do it , whould i store them upside down?
Going to buy a blade and a torch

Condensation won’t hurt unless it pools up over the myc.

I either leave them unwrapped in the SAB for like a week or wrap them and stack them on their sides until it all pools up and then open and dump it in the SAB.

I’ve seen people wipe it with a paper towel but I haven’t been that brave yet.
