I almost bought a separation funnel a while back just for this, but it was an extra step to have to strain the root bark out each pull and then put them back in. In retrospect I suppose I could have done 4 pulls then put bark back in but I liked them soaking as long as possible. I did the baggie separation funnel back in the day extracting DXM. Lol haven't thought about that in years. The 10ml pipette was a HUGE improvement. 50ml pulls in only 5 squeezes of the pipette instead of a fuck load with the 1ml ones.
But I digress from the topic of the awesome thread and don't want to lose focus. I have been held back more than expected but for really reals should get some transfers done today. The up side is PC'd about 26 jars and 3 bags so I'm gonna take a chance with some and throw some early transfer wedges in as well on making T2s and T3s. Should also finally get my Rusty Whytes started and make my T1 of Z. May go ahead and take a chance with that on and throw some in a pt jar. Lots of updates\photos\video coming.

Ok I need some help here, putting agar to work for the first time. I dropped this through the grate in the SAB trying to cut the stick off and none of the plates I swabbed are doing anything. It looks like I can get a clean transfer around 9-11 in the first pic.
 
Looks like that’s the spot grown furthest from the swab and furthest from any visible bacteria. What’s the play here.

I never get anything from swabs. I either go loop to print, or I shred the swab fibers with my scalpel and put in the plate. From there I hope that it's as clean as could be, otherwise its Seal Team Six time going on a rescue mission.

I agree with the 9-11 o'clock on the first pic. Go for just the faint bit on the edge and only a tiny amount. If you have enough plates, just 2 or 3 tiny bits. The smaller the transfer the less room for contams to come along, and even the tiniest bit will grow.

I had read other people sayjng that and initially tried to pull fibers off with tweezers but they were just too tough.
Originally I buried the swab in another plate and it was totally covered in bacteria, then myc sprouted from the top of the swab. So I took it and dabbed it around this plate to see if anything would take before sticking it in the middle.
That’s what all the bacteria spots are from.
I really hope this works I’ve been looking forward to melmak.
Also I can’t tell but I think that light halo which is more visible over the swab in the second backside picture is from the swab lifting the agar off the dish. But it could also be bacteria.

Perhaps I got lucky but my first attempt with a swab went better than a loop. Growth along the zig zag pattern almost perfectly. Germed in 3 days I believe.

I've definitely had far more luck with swabs than I ever did with a loop... I hated the loop from the start... lol

I think swab is a little easier, but I’ve looped enough now to not mind. My main concern was not knowing if anything was on the loop.
For some reason the agar I used on the first is just horrible for spores. Still no growth on any of them, but I made new softer plates and swiped the same prints the same way and they all went off on day four.

Are the people actually working in labs not using glass petris? Or...to avoid massive condensation, plates have to be poured after autoclave. Is anyone in the industry using glass? Perhaps it's all disposable plates now. My illusions are shattered!

So never having done spores to agar before, I keep having questions.
Is this far enough along to take transfers or should I wait until it fills in enough to see sectors or whatever?

I personally would take some from the edge of the 7 o'clock bit and some from the 1 o'clock bit. I also usually let those plates keep going and try to take more later on.

This is my first big adventure in agar with my FH and I did 2 swabs and got nothing so far on those plates... but the ones I scraped/looped are doing good. I'm going to stay away from swabs for now.

all disposable petris ime
back when it was my job I even made all my small batches of LCs in reusable autoclavable plastic bottles. its reusable, but glass is expensive and breakable too. with the number we went through it would have been cost and time prohibitive.

This is one of the spore plates. This was the first time I used a loop (DIY and low effort but it worked!) and did a zigzag pattern, and got decent looking growth.

I took 3 transfers from each plate (PESA and TOC) and plan to let them grow out a bit before further transfers. I'm waiting on a current batch of jars finishing up so I can get them back for this project, so no rush yet.

Im planning to use WBS in pint jars (well, 500ml jars and thats still 68ml short of a UK pint, I guess for this hobby its mostly "live as if" when it comes to imperial vs metric measurements.) Quart jars don't fit in my PC. Also I've been having success with using jam jars and pasta sauce jars so I'll be saving up more of those.
What size grows are peoples planning on doing? I can only really do a couple of "quarts" at a time which isn't terribly exciting, so I'm hoping to create some novelty grows to make things more intetesting.

Tattersail I'm probably doing shoeboxes but may do some g2g for monos if the jars look exceptional.
RW and GT T1's going to 2 quarts each of rye today. Not as uniform as I usually prefer but the germ plates still look good and I'm leaving for a 7 day vacation in a few days. Pan Cyan and Natalensis will get another transfer.
