500 Petri Dishes, how many do you typically pour at a time?

I'm getting sick of working with no-pours and am very tempted to pull the trigger on a 500ct of petri dishes and get back to these instead of mini rounds. Just out of curiosity, those of you who do extensive agar work, how many plates do you typically pour at a time? I hate pouring in the SAB, but I think I prefer pouring in the SAB opposed to dicking with no-pours.

I normally poor 20-40 dishes in my SAB at a time depending on what projects I have going. I normally have to poor agar 2-3 times a week. I just built a flow hood so I may start pouring more at once so I don’t have to so often.

20 in a SAB. As many as 100 infront of FH.

Dude the case is so awesome, do it.


I usually do 20-40.

But now that I have more media bottles I’ll prob go for 40 minimum each time.
Depends what you’ve got planned too. I’ll just pour a sleeve, let it cool then put them back in the sleeve and back in the box til I need them.

A sleeve holds 20 and Bodhi says that's the right amount for a 500ml media jar filled up to 400ml. I usually end up with about 24 and probably need to pour some a little thicker.

Pouring 20 is very convenient because they fit back in the bag, and also so you can finish while the agar is still liquid. I was pouring at 120 degrees and then 118, but I'm going to try it lower next time and try to reduce condensation. Bodhi's video has a system of pouring large stacks from the bottom but I tend to screw this up, and am looking for other ways to reduce condensation. Putting a flat bottomed cup of hot water on top of a plate is supposed to work but I haven't had much luck with that either.

Thanks for the input guys. Thinking I'm going to pull the trigger. Before I was using grafting tape instead of parafilm. Would you advice against that, or either is fine? Plates seemed to grow just fine, the tape would break or not stick occasionally, but for the price it seemed like the way to go.

Ill pour one sleeve at a time, maybe more if I anticipate needing them.

When I sterilize agar though, Ill do mulitple bottles, then just warm them up in the microwave to melt them down for pouring.

the sleeves in that case are 25. I usually do 2, 500 ml for each sleeve for 20ml per plate. inevitably I end up pouring some thinner and some thicker. no biggy.

Yeah I cut them with a box cutter. It was way easier than I thought too. And now it’s super fast to wrap the dishes and you can see them way better.

I thought it would be a pain in the ass so I was just tearing off like an inch and a half wide strips for a while.

I usually pour 20-40. recently I've been upping it to see how much I can handle in my SAB. I also like trying to pour them a little thinner if I can, just to save on ingredients. My last batch was 1500ml (between 2 bottles) and ended up pouring out to 78 plates :eek:

Wasn't too bad actually, could definitely use a bigger SAB though.

My advice is get small media bottles and use multiple for big batches... 500ml bottles are way easier to pour than 1000ml bottles.

I make the PC cycle worth while and do 3 liters of agar and pour like 50-60. I like having and using old plates because if they don't have contams in a month of sitting you know you're looking good.

I like to bulk pour. I have three 500mL bottles, make two PC runs, and pour 120-150 plates. Thin pours, i can get 75 plates from three bottles. No such thing as too many blank plates laying around! :chugbeer: I'll probably slow down eventually, i'm just working with a dozen different cultures/varieties ATM.

Great info, thanks again everyone for the input. I think its decided, 500 petri dishes coming up. I just bought that 3 pack of questionable media bottles off amazon, so well see how their quality holds up. If nothing else I have 1 nice 500ml Pyrex bottle I can use to knock out a sleeve at a time. Bought myself some Cling Wrap last night :rockon:

Condensation is a matter of external and internal temps being different.  I pour my plates, wrap with cling wrap, then store them flat in a ziploc bag.  The condensation goes away overnight.

I had condensation problems when I poured two weeks ago. Poured at 118f but still got it bad. Looking back though there was a period where they weren’t all kept in their stack of 10 because I was having confidence problems lifting up the lid on the bottom two plates with the other 8-9 plates stacked on top of them. Was too scared I was gonna tip them over and ruin everything so for a short period (the duration of the pour) they were in stacks of 5. I tried the hot water cup on top the next day and it just melted the parafilm.

I poured a few days ago at 118f but kept them in the stack of 10 the whole time and then right after I poured the last one I set the hot cup of water on top of the stack and I have no condensation on 9/10 plates. When doing the hot cup of water on top of the petri make sure you’re using a metal cup (I used a small sauce pan the first time before I found something better)

Both times I poured at 118f and both times I only did 10 plates and both times by the last plate my agar was getting chunky. I wouldn’t go much lower than 118f unless you’re only pouring 10 plates too or unless your residence is kept very warm. Or do what I did and make about 100mL more agar than you’re going to actually need because it reallly starts cooling down once you start pouring it because there’s not as much in the bottle to keep itself warm. So if you make about 100mL extra you should be able to pour them all just fine because all the chunky stuff will sink to the bottom of your agar pour dish leaving you with enough liquid agar to finish the last plate

Are you storing them at room temperature? I've been putting them in the refrigerator. Maybe that's the problem.
