Agar plates, not sure whether contam.

Hey,

This was a first attempt at Agar and I was wondering if anyone could help tell wether these are contaminated or not? All assistance is appreciated.they were inoculated using an MS Syringe and the growth doesnt seem as radial as I've seen but I'm not sure if that's due to over solution or is typical of a MS.

Edit - just to add they're around 3-4 weeks old.

These don't look contaminated to me (the one in the last picture looks a bit questionable). But they look overgrown to me (for transfers at least you don't wait for full petri colonisation) - depending on what you wanted to use these for.

These look pretty common to what I'd expect from a MS inoculation - lots of separate genetics sectors from spores. For the radial growth you described you would have to transfer parts of these to a clean new agar plate (could work now but ideally should have been done week or two ago it seems).

you’ve definitely got some mycelium there. the plates look a little overgrown but they look clean to me. it’s a little hard to tell from the pic though, there could be bacteria or something on them. if you see milky clear stuff on the areas that aren’t covered in mycelium & aren’t pure agar there could be contamination. if you used a MS syringe from a vendor & practiced good sterile techniques they’re probably clean. i haven’t personally made a plate from MS solution, but i’m thinking that maybe the spores got moved around from the water or something & that’s why you don’t have the radial growth you usually see on agar. looks like a lot of tomentose growth there as well. not sure what causes that on agar outside of genetics, but i’ve had plates that look like that too, especially from spores.

if i were you, i would transfer a wedge to a new plate just to assure you have clean spawn, as there could be hidden contaminates under all that growth & it never hurts to be certain. however, if you really don’t wanna wait for more plates to grow out you might get lucky spawning those.

congrats on the agar work! once you get a feel for it it’s super easy.

lol i had my reply typed out but didn’t get a chance to send it until just now... guess you’re looking decent OP. you should be fine transferring those plates as is, but yeah typically transfers are at the first sign of clean growth.

That radial growth you're looking for only happens after you do your first transfers.

There is a lot going on in those plates, and its going to be hard to pick out any funkiness. You should do a few transfers and then go from there.
