Do you clone from the stem or three cap?

Hey all, I am wondering what people's preferences are for cloning on whether you close from the cap or stem, and why?

Typically I tear the mushroom from the bottom of the stem up and take a transfer from inner tissue  close to where the cap and stem meet. I don’t like taking directly from cap to prevent germination of spores I don’t want.

I’m pretty new to agar but I’ve taken a bunch of clones. Always torn open from the bottom of the stem and cut tissue from the middle area.

Only reason being that’s how I read to do it.

Good point about rogue spores when cloning a cap, that would defeat the purpose.

I love the cap.  I take clones before the cap opens, I hope its not yet sporulating then. 

Tear in half, up from the bottom and there is such a nice, thick spot to take a sample in the centre of the cap.  I just find the stem is harder to work with.

The cap and the stem are both the same mycelium. Taking a sample from the cap or stem should be genetically the same :shrug: or no?

It's genetically the same but the composition of the cells isn't necessarily the same. It doesn't really make a big difference though. In mature mushrooms I usually snap the cap in half and cut a piece from the center near where the cap meets the stem. I prefer to just clone tiny pins and put the whole pin on the agar.

Maybe I am wrong but I think dropping a pin onto agar can be ideal when going for most healthy aggressive mycelium; but really works best if you are working with a known culture with a stable, examined morphology and tested for potency. :shrug: If you are going for a mature fruit in a new grow with huge disparities in genetics, the argument of spore contamination could be made if choosing to clone by taking a sample from the actual cap full of spores.

Its the same as far as I know, I just find the actual mechanics of taking a sample  work out better on a cap than a stem.  Deeper flesh.  Firmer denser and easier to work with

If you do go for the stem, you gotta watch out for that ropey, centre mycelium running up and down in there.  It won't grow.

Wrong. Any mycelium from the fruit body will continue to grow.

That works. Most often I'll put some spores on agar and if there are no visible contaminants let it grow out and start pinning and use of those pins. It's such an easy way to get an isolate you know will fruit with little potential for hidden pathogens.

No.  A monoculture take awhile to obtain, according to these boards.  Even cloned mushrooms are more than one strain, if you can believe it.

I just make sure to be as verbally abusive as possible to whatever sample I’m taking from whichever section of the fruiting body; you know, like most boss’ treat their employees these days. Good for morale.

Sounds like a good time for me to try this since my agar petri are showing zero signs of life. I’m certain it isn’t the agar... or is it? Only one way to find out...

I've never cloned before and was wondering if there would be any advantage of "washing/rinsing" the specimens first in say in alcohol/iso first to help get rid of contaminated hitch hikers or is it just done on transfers to work away from the baddies? Was obviously trying to make less transfers if possible.

You can wipe it down gently with an alcohol soaked tissue if you want.
