YogiBear's No Pour Agar Tek
( this is a flow hood tek )

⚜️  THESE PP5 PETRIS ARE TOTALLY REUSABLE  ⚜️
⚜️  SIMILAR TO REGULAR PETRIS IN SHAPE AND FUNCTION  ⚜️

👎  THEY DO FADE A LITTLE OVER TIME BUT NOT BAD  👎
👎  THE LIDS DO WARP A LITTLE  👎





⚜️  YOGIBEAR'S AGAR MIX ⚜️

Hanna Magnetic Stirrer (2in stir pill)
4 cup Pyrex Measuring Cup
250ml Cold Bottled Water
Add 5g LME
Stir 2-3 minutes
Cloudy -> Microwave 2 mins on high
Clear -> Stir in 5g Telephone Agar
Add 3 Drops Food Color
Stir well and Pour 12 plates (20cc each)






⚜️  WRAP 2 PLATES WITH ALUMINUM FOIL  ⚜️





⚜️  LOAD IN THE PC  ⚜️
( 24 fit easily into a 16qt Presto )

⚜️  PC 30 MINUTES @ 17.6393645 PSI  ⚜️





⚜️  LET IT DROP TO ZERO  ⚜️


⚜️  TURN ON FLOW HOOD  ⚜️


⚜️  COOL, WIPE AND BAG UP  ⚜️
( lay out to cool in front of flow hood )
( quick wipe on lids for moisture w/paper towel )
( a new paper towel is sterile, I use 1 per plate )
( individually wrapped in sandwich bags )
( OR use para-film or Saran Wrap instead if you'd prefer )





⚜️  CLEAN AND CLEAR  ⚜️





⚜️  HOUSTON... WE ARE READY FOR LIFT OFF !!!  ⚜️

AND REMEMBER TO RECYCLE AS MUCH AS YOU CAN
ALL MY FOIL, PAPER TOWELS ARE USED ONCE
IN THE LAB THEN THEY GO TO THE KITCHEN

Yo that's pretty cool and all but I'm all about pouring plates these days...

Sandwich bag thing seems sketchy tho, all that air in the bag is contaminated and those lids on those plates are loose AF...

Oo ok ya then in that case no biggy as those baggies are manufactured with molten plastic and therefore be sterile on the inside...

So that's the only thing I could see being an issue..

Sweet idea tho... if i was doing no pours I'd much rather do these just for the sake of the clear lid.

So if you'd buying petris and all then why dont you just pour sterile plates? is it because you wanna be able to reuse them or  something?

Side note: after realizing your a grandpa and now seeing how you are able to make your on GIFs and shit I'm seriously flabbergasted...I thought grandpa's and grandma's were supposed to have a hard Time even looking at a smart phone or computer haha...  dude I dont even know how to do that shit!

Great write up - I always appreciate good quality pics; hope this helps someone out here. Don't take it the wrong way but the first thought that popped in my head when I saw this TEK - does he/she use it themselves even if they have flowhood available? That should make pouring agar with close to 100% sterile as possible :laugh:


I have just recently started with pouring (not tried non-pours ever) because I was getting annoyed just by thinking about individually (or in packs of 2 or 4) packaging plates in sandwich bags. For bigger quantities or higher plate turnover rate this seems like a lot of busy work.

While I'm still far from perfect sterile technique - I prepare a large media bottle in advance and keep it in the fridge, then I microwave it and let it cool down for pouring - can get 20-30 plates done in a few minutes with a good success rate. Then I can store them later - then cool them down while still stacked in the SAB and put their 20 stack bag packet back on for storage (or use right away).

At the moment I'm at the "eager agar" mycology stage when I want to see and try  lot's of stuff so I go through plates much faster than I would like to prep no-pours.

I have had jobs as a web designer, book publisher graphic artist and designed ads for one of those companies that sends coupon mailers to every house in America. That's where my eye for design and software comes from.

Also lots of network experience and network protection for old school internet service providers.

I only use iPhone for image work...

I do such small batches so I like the no pour tek the best

If I had a lab then yes, I'd do normal pours.

I could just tilt and dump the moisture out onto a paper towel but I want to get as much out as possible so I just pop the lid up in the flow hood stream and use a fresh paper towel folded up to about 2in square and use the edge.

Definitely use a new paper towel for each petri 👍

So are these reusable? That’s a great price. Do you still have to wrap with parafilm or saran?

Also why 17.9 psi?

Yes these PP5 are reusable Eats and the 17.927456 is a complete playful joke my friend 😎. I guess I should edit the OP to reflect that.

I only use the sandwich bags because my hands are way to shaky to work with the para-film and wrap. I can just pop open a new bag in the flowhood stream, drop the petri in and done.

They are like regular petris and would need para-film, correct.

I gotcha.  Well, do you keep your paper towels in a sterile environment or just sitting on the counter? 

Wouldn't they pick up contaminations sitting in open air..?

My body piercing room was tiny with no air flow when the door was shut so I would have to assume from that and the State Health Inspectors statement that's its both the room and paper towels being sterile at the manufacturing process. She just said "use fresh towels" for each procedure/person/client I had.

None of my customers ever got infections.

As the pictures above show after a 3 day waiting period... no contamination.

BTW I use proper Flow Hood technique and keep the lid I'm wiping closer to me when I wipe so there is no chance of paper towel "stuff" getting in the plate and always let the air flow blow on the inside of the lid for a second or two before putting the lid back on.

I will be making animated gifs for this procedure but I don't have a way rigged to hold my phone. Usually have one of my kids hold it for me and they are all still on vacation right now.

I've been using these for a while now. I bought the 75 and 100mm dishes. I just toss them in a big ziploc container with a vent (covered with tyvek and MP tape). I was positive they'd boil over but they all came out perfectly (of course not 100% if you count the condensation, lol). I've had some dishes sitting for over a month and saran wrapped with not signs of contam. I use about (I think) about 8-10ml agar mix in the 75 and 12.5ml in the 100mm.

I still have some prepped and laying around but switched to 4 and 8 oz mason jars because of how easy the agar just comes right off of them. Using SAGARI tek the agar sometimes sticks to plastics a little too much and makes it a pain.
