Lion's Mane Clone on Agar

So I foraged my first lion's mane the other day and am trying to clone it.  I put a couple small pieces of the inside meat on some MEA agar two days ago and do not see any growth at all.  Does lion's mane grow slow or should I be worried.  Are there certain types of agar it prefers that I might be able to transfer it to?

My lions mane is pretty slow. The mycelium can be pretty thin and hard to spot.

I completed a transfer ten days ago and at first glance it looks like it didnt take. If I get in real close with my glasses and a headlamp I can see it has actually got some thin mycelium spreading.

I'd say give your culture some time.

I currently have some LM plates that busted a fast wide groove the next day after inoculating the plates.. but the fruits grow slow as you know what for this one and when it was colonizing the spawn bags it did it slow and looked forever like nothing was happening... but drop it on the plates it is so far fast as a rabbit. :shrug:

What kind of agar do you use?

Currently I am using BRF agar. And I didn't strain anything out. Its goopier. So I might not do it that way next time.. I haven't fully decided yet.

I've got my LM on BRF agar right now also. I'm going to cook up some MEA today and see if it performs any better.

Of course, my cat keeps opening the door to my heated room so things may just be slow for me because the temperature keeps falling!

Hopefully it's not too late, but from my experience lions mane on straight LMEA is very slow. It would be best to throw some sort of nutrient in there. Potato flakes work but nutritional yeast would be better.

Good to know..yeah it is showing zero signs of growth still.  Just sprinkle some yeast on there and it might help?  What about making a different kind of agar and transferring? It's been off the tree over a week now..I hope I don't lose this clone. It was one hell of a fruit.

No, do not sprinkle yeast on the culture now, it needs to be added during agar medium preparation. Also it's not bread yeast if you didn't know. Nutritional yeast is a cultured and dried yeast that contains various amino acids, minerals and vitamins that are beneficial to mushrooms growth. If you are really concerned you can transfer to new medium, but from my experience and depending on the source of the culture, it can take 3-5+ days just to show signs of growth. Be more patient and when you see first signs of growth transfer the tail end of that new growth to new agar culture. To do it this way is supposedly recommended and common practice anyway, it ensures a clean culture.

Also it might help to see the growth by using a strong flashlight and shinning it underneath the dish and looking at it from the top. Lions mane can be very hard to see when it is just starting, when it gets old it starts to thicken up, but before then it's pretty wispy and strand-like.

I've had no problems with Lions Mane on MEA. It depends on the culture... one of the three I have is vigorous on MEA and is even starting to fruit off the plate. That said, as already stated by others, if you are having issues with slow growth you may want to try PDYA or something with a bit more nutrition.

I have recently been doing 50% grain water agar with an added 2-5 g of MEA per 500 ml and it does well with all my species-

LM tends to grow in 3 D on agar in my experience so appears slow, but really it is just growing down as much as out.
