4 year old spore swab, any solutions?

I have a spore swab thats around 4 years old or so. I streaked it to a couple of plates, but a month later and there's no growth on the petri dish whatsoever. I've read some stuff here and there about forcing old spores to hydrate by making a spore syringe, capping it with a plug, and then, using rubber bands to apply pressure to the plunger, put the spore solution under high pressure for a day or two.

Thinking about cutting the head off the swab into a shot glass of sterile water and sucking it up with a syringe then putting it under pressure. Does anyone here have any experience with ths? Any success stories with old spores?

How were they stored? Were they kept dry? Ive never used a swab. Id rehydrate them and try again. Idk about keeping them under pressure. Never heard that

Kept dry, sealed in a ziplock inside of a quart jar in a desk drawer that was opened a few times over the last few years. Its how Ive kept all of my spore prints and swabs, in their own bags inside the jar. All of my prints are 4+ years old. Ive only had luck getting one print that was around 5 years old to germinate on agar. Everything else Ive put to agar hasnt germinated at all

Id rehydrate them. I think if you store them in the fridge they last longer. Ive heard of people laminating them also

Exactly what I was referring to. Im just worried with a spore swab that the spore solution made up will contain contams. Its all going to agar but my fear is with a solution all the spores and any present contams will be mixed together

Almost everything I'm growing rn is 8yrs or older. The hydrating thing didn't work for me on at least 4 prints but maybe it will for you. What I have had luck with is being generous with the spores and going from print to agar. If I don't see germination in 3 weeks I trash it and try again using increasingly more spores. Some of my prints do seem dead but lots of others just didn't germinate on the first few plates, persistence paid off.

Creating a spore syringe isnt the issue here, did you read my post?

Word. I may have to just scrape more spores off my prints onto the agar dish. I usually apply as much will fit on the end of my scalpel.

I've made 3 attempts so far with the spore swab in the OP. All 3 attempts of growing it out on agar resulted in 0 germination. I cut the head off the swab today into a shot glass and made a spore syringe from it with sterile water. Going to give the forced hydration a try, let it sit for a few days for the spores to hydrate, and drop some of the solution onto a few plates

Recently used a print about 5 or 6 years old... Swabbed 6 plates, only one germinated...  Took about a month before I saw anything.  I needed to use a generous amount of spores...

Tried rehydrating in syringes but had absolutely no luck.

Saving cultures is one thing but fighting spores is hardly worth the battle, they are easily obtained fresh

Just trying to work with what I have. I had a decent spore collection piled up 4-5 years ago, quit growing and recently got into it. Im in one of the 3 states that you can't order spores online to. There are other ways to get more spores tho :shrug:

Yeah swab it to a plate of agar. I did a 12 year old print with success!

Thats incredible. I had 2 swabs. Tried both of them a few times on agar, each time threw the plate away after a month of no growth. I was able to get some mazatapec spores that were around 4 years old to germinate via spore syringe (This was prior to me getting some agar supplies, was just getting back into growing.) Now I have plenty cultures of the Maz which Ive been working with, but all my other spore prints/swabs Ive had zero luck with germination. The Maz just happened to be a miracle apparently
