Questions about beginning with agar

Hey so I'm finally about to begin cultivating for the very first time and I'm just procuring materials to start with.

I'm just confused about the following:

1)What materials will I need for proper sanitization? Iso Propyl Alcohol or Ethanol, which works better? Do I need to buy things like bleach?

2)Further with the Agar recipe, which should I go for? Is PDA better or MEA?

3) What is the parafilm for? c10's agar tek tells me bod uses saran wrap. But I've also heard people say you can use micropore tape. So I guess I'm wondering if the agar plates need gas exchange or not?


These items aren't actually cheap where I live, and the alcohols are hard to procure as well. In fact I'm just working with 8 petri dishes for now. So just making sure before I go ahead with the purchases.

I like to use a cling wrap roll cut into 1 inch sections in place of para film or micropore tape. Tape is the worst. It works fine but is a bitch to get off when it comes time to transfer. I like MEA cause it's stupid easy and the 1# of LME I bought a year ago is still half full. Best advice I can give is to get a case of Petri dishes and start doing it. You'll figure it out as you go.

Oh and you don't need bleach. That shits bad for your lungs and will fuck up your clothes. 70% IPA in a spray bottle, gloves, and a mask are all you need to pour plates. And an SAB of course. Then you'll need a torch and exacto knife or something comparable to make transfers.

Agar and LME are cheap. Petris cost a bit. Just remember to keep your LME in a mason jar or it will turn into a rock. And order one of those 500ml media bottles with the orange cap from Bhodi's agar tek.

If you're worried, try a no-pour tek

I've had great success making MEA by mixing in a small jar and submerging that jar partially in a pot of boiling water to melt it up, then pouring into glass plates. Then wrapping the plates in stacks of 3-5 in a couple of layers of kitchen foil and putting them in the pc for 20 mins.

After unwrapping and inoculating in a SAB yeah i find cling-film works great to keep them sealed. GE isn't really that much of a problem because the mass of living mycelium compared to the volume of air trapped in each plate means that it has enough oxygen to do it's thing.

Sorry I didn't read this very well and have now had some coffee. I see your problem. So you got glass plates and only 8 of them or you got some plastic dishes? If they are plastic and not sealed well they may be no good. you may want to look into no pour agar if that's the case. You will need access to 70% IPA. There are some folks working in flow with no gloves or alcohol but that's after years of practice. I spray my hands down Everytime I notice the gloves are dry and anything that goes into the box gets sprayed first. I use a 1/4 bottle per session Wich is probably Overkill but I got clean cultures so I don't plan on changing anything.
