Just wanting to know if any of these are worth a shit. some have multiple syringe drops obviously. It was My first attempt and I wanted to make Sure I got some spores on the AGAR. I also inoculated some oats from Syringe and everthing is taking for eV er. Really starting to think the humidity may be to Low in the room in working in. It's around 25-30% Thanks fam

Wow those are hideous. I see some mushroom mycelium but its either on top or right next to some freaky shit..
I would do some new plates and swab the plate in a z pattern or dab it in the middle... you have no way of knowing what went where..
It seriously looks like u inoculated those in open air too...?
What was ur technique like?

Try to only use 1 drop of spore solution. Rooms RH isnt gonna be a factor.

Hideous is exactly how I feel about them. That's the confirmation I was needing.
Btw did it all in a SAB. However I forgot to wipe the dishes with alcohol before opening and I'm not 100% sure my PC process went as it should have.
Shitz I've never tried the medicine and the wait is driving me crazy. On top of that I'm not the most patient person in the world but I do like a challenge.

I wouldnt just squirt spore solution in your plates... definitely would take a loop or a sterile swab and put a drop on that and then touch it to your plate...
Also I dont know if you knew cus it looks like you took a pic from directly above your plates but nothing other than your sterile tools should ever go above your open plates...
Next time save a plate and dont open it and see if it contaminates then you'll know it was something to do with sterilizing process...
Also please dont dump your plates cus I said they look gross lol maybe someone with some more experience could guide you through a couple transfers...

With a dirty syringe its best to use a minimal amount of solution and spread it around thinly as mentioned above, a zig zag down the center of the plate is a good pattern.
There are streaking patterns for super contaminated samples but yours while not clean doesnt look like it would need that. (Essentially you spread along one side of petri, then flame and spread from that site up to one side and then flame again and spread it more and l work it towards the center of the plate eventually. Look at the agar link in my signature for a diagram)
Its also best if you can get a transfer away from the germ plate asap I pike to do transfer when the growth is about the size of a nickel or quarter ~ 1β. Often there is very faint mycelium at the leading edge, if you can get a rice grain sized piece of that youβre usually golden.

So do any of these look worthy of a transfer attempt or should I just start over? Really dont mind either way. Just want to do It right.
Its apparent to me now that I probably was not taking the sterile environment part near serious enough. I have alcohol, gloves, SAB, lighters...etc. Im in construction so not quite used to the careful and clean techniques yet.

I think the main contam is bacteria from the syringe. I see yeast in 5
4 or 6 would probably have been good for a transfer 1-2 days ago, 1 maybe 2-3 days ago as well.
You can try from 4 but its already grown up close to the edge so it will be hard to catch a true leading edge. Cant hurt to try a couple from the more organized looking edges. Worst case it transfers dirty and at least you have some more space to try to outgrow the contams.
Iβd also start some new plates either way though. Always good to have backups for your backups.

I understand that transfer from the leading edge is best but if you transfer from the middle of growth wonβt it still grow? I am asking to learn.

It will still grow but it wont be any cleaner. Thats why I was saying you should try and catch an edge but if not youβre basically just buying some time/space for it to grow away from the contams
And yes but those also look bacterial just like your plates so idk how much it would help you really.

Imo I think plucking a grain would be easier than trying to do a transfer from those plates...
If he played his cards right he could probably get a clean culture after a couple transfers from a colonized kernal...
