Spores On Agar Visual Question

Hi Guys,
I attempted my first spore swab to agar 5 days ago. I'm trying to work out if the bigger spot on the top left of the photo is healthy mycelium or a contam.
In the right lighting it kind of has a shine over it.
Was hoping to get your thoughts?

Bacterial contamination, and you are unlikely to see growth from non-fresh spores after 5 days. I waited 2 weeks for my first mycelium to be visible.

Let it grow out and you will hopefully be able to tell what’s mycelium and what’s not

Still kinda hard to tell. I think the top left spot is bacteria but it does look like there’s some germination on the plate. Give it a couple of more days and you will be able to tell.

So did you use an inoculation loop? Does that line pattern on the agar follow the streak pattern you used?
If so I'd grab something quick, I"m thinking the 5 to 6 oclock region, and do a transfer.

It was a spore swab. But yeah it 100% follows the pattern I followed.
You reckon do a transfer now or let it grow out some more?

Since spores are dirty from the get go I'd recommend grabbing some of the best growth straight away and transferring to a clean plate. You want to isolate that clean mycelium. If you go straight up from 6 oclock there looks to be a good rhizo cluster to grab.
I'm sure others have opinions to offer and take mine with a grain of salt.

I would make transfers now. For plates like this, its best to post some pics with light shining through the back of the plate too. It helps to identify contamination.
I would avoid those satellite colonies at 8 and 9 o'clock.

More than likely it is not killed off and it is now hiding with your mycelium. Don't transfer from the area in question. I use a sharpie to mark bad spots so after you get growth you know where to transfer and where not to. I will X bad spots and O good spots.
