I read a couple pages CBK linked about those Holy Grails. What was the outcome, for preferred PC method? Did quart jars prevail?
***edit*** I read the rest of the thread. I'll keep an eye out for more testing. I'm def getting some next order.
And I also had trouble seeing pics. Link to titties please.

update 1/12:
this was all mold except the oyster. "lol"

so for now the King Oyster will be the only gourmet-from-spores I'll be doing from that batch. I just picked up another mixed pack though and it's got chestnut/velvet pioppini/whatever you want to call it in it so, why not try and grow those too? why the fuck not. throw it in there. maybe tomorrow.
tonight I'm prepping coco for PESA shoeboxes over the next few days, starting with the HEAVY jars which should be ready to go come AM. I love LC because it just goes and goes looking like normal ass grains, then BOOM! myc all over.

Got tired of waiting for plates to germ some did up some PDYA in no pours at 3% nutrients (made sure I did my math right this time by calculating based on DRY nutrient weight )
Swabbed some pan Cambo var sandoz, pan Bisporus (HUGE shout out to mary for the kickass prints )
Swabbed a couple RW as well, OP was updated

January 12th:
Finally back from vacation and all my plates are ready for their first transfers:

Going to be an interesting time with these. The pans are just new to me, so I'm mostly just guessing at what looks good. The PE plate will need working around that bacterial spot. And the B+ plates are all muliti-spore as fuck - I only had soft agar plates laying around and the loop sunk right in, so they didn't get spread out much and there's a ton of overlapping growth.
(And don't pay attention to the weird coloring. Evidently myc loves eating the blue food coloring I use but not the red or yellow, so the agar changes color as the plates colonize.)
Wish me luck tonight...

I spent the first 11 days of 2020 on vacation, so hadn't had a chance to do any myco work this year until today (though I have been following the thread). I appreciate CBK encouraging those of us who couldn't start on the 1st to participate anyway. I really hope to learn a lot--I have only once before inoculated agar from a spore print.
Here's my first update copied from my OP:
Quote:
January 12: Was home from vacation for less than 24 hours before getting spores to agar. In an interesting twist, my unheated garage (where I store my myco gear) froze hard while I was away. This includes about the half-dozen spore syringes and half-dozen spore prints I had on hand. A quick search of the Shroomery forum turned up conflicting speculation on whether cubensis prints would be destroyed by freezing. Most people seem to think that spores suspended in water would be destroyed by freezing, but prints should be okay. I'm going to try to use the Koh Samui and Mars prints, as originally planned, but I'm also going to try a Transkei spore syringe (after thawing) just out of curiosity. I'm going to be pretty bummed if all of these spores are ruined.
Used some agar plates that I poured about a month ago using a recipe of 400 mL of water, 6 grams of LME, and 8 grams of telephone brand agar-agar. If I was going to make new plates for germination, I would probably have reversed that LME/agar-agar ratio, as the last time (first and only) time I tried scraping spores to plates they took a week to germinate. But these were what I had on hand.
Set up my SAB with a wire rack on top of four half-pint jars for height. Sprayed and wiped all tools with 70% iso. Sprayed the inside walls of the SAB lightly with soapy water. Took a quick shower and gargled with mouth wash.
Used a disposable inoculation loop to scrape up spores from prints, then spread on agar plates in a "z" pattern (from what I've been reading of this thread, swabs are more popular--I'll try that someday but have no sterilized swabs on hand at this point). Initially inoculated three plates each with Koh Samui and Mars spores. Switched loops between each print. Flamed loop on gas range between each plate, cooling in receiving plate. Inoculated three more plates with a small drop each from my Transkei syringe (shaking the syringe before starting and flaming needle tip between each pate).
At this point I realized that I had been spraying my hands and arms with SOAPY WATER AND NOT ALCOHOL. So, after dousing myself with alcohol for real this time, I went back and used a new loop to scrape spores from the Koh Samui print to the two plates I had left.
Between the frozen spores and the poor sterile technique, I'm definitely a little worried.

January 13th:
First transfers complete:

Public service announcement: if you decide to be more eco-conscious and get some glass petris to try out, don't try to use them as no-pour plates (meaning, don't PC them with agar already in them). The condensation and general residual wetness is a pain in the ass to work with, and definitely adds an extra potential contamination vector - hopefully it doesn't come back to bite me too hard. Lessons learned, I guess?

What? My dreams shattered, glass petris are not PC'd with agar?

I have been thinking about getting some glass plates. At this point though, I think buying enough to not need any plastic plates would be prohibitively expensive. I'm about to start flying through some plates. I really hate throwing away my plastic Pettis though for multiple reasons. Which\where glass petris did you buy?

Hey there Durgin. I do no pours with PP5 petris (in my signature) and do a quick wipe on my lids when they cool in front of my flow hood with no contamination. Maybe this could work for you?

It can be done, but the problem is not only extra water on the inside but also on the outside and between the edges. Basically it's just a mess. Not only is it ugly to look at and a potential contamination risk, but the wetness makes parafilm not stick well.

These are the ones: https://www.amazon.com/gp/aw/d/B07RP21ZRS
I think they'll be ok with oven sterilization and a regular pour, I had just hoped to avoid that. Also note that these particular ones have a really loose fit and total overlap (so the gap is on the bottom), which makes wrapping them harder.

That could work with a hood, but definitely not a SAB. The water in these is also between the edges though, so it's more work and contamination risk, definitely not ideal. I'm just going to go back to regular pouring - I'm pretty solid at that (maybe 1-2% contam rate in my SAB) so not a big deal.
May need some of those pp5 plates though when they're back in stock. I've quickly figured out why more people don't use glass, but at the same time I do prefer reusable supplies - I don't have to feel guilty about taking extra transfers when it's just a little extra agar.

Yeah, just bought two packs today. I'm just using the reusable plates for short-term transfers, still sticking with disposable for long term master/slave plates (+ slants).

Did a couple 2nd transfers today. 1 RW T2, 2 AC T2
A prior PE6 being cleaned from MSS. This is the prettiest I've seen yet and a monumental step for me.

here we go

I kept the sub a bit on the dry side for the "heavy" box since there was so much LC in there it's certainly got enough moisture. spawned to coco at 1:1.5 with a .5 casing, so I labeled it 1:2. gave the casing a good mist and closed them up. my last run like this I harvested the first flush yesterday after 15 days. if all goes according to plan, I'll have some PESA by month's end.
