Also I remembered that I ordered a spore syringe sitting around. I happened to have a dish in my batch that I forgot to swab and appears to be clean, so I might try footpaths "drop & tilt" method for fun.
I actually use a paper clip and an Xacto knife handle for my noc loop. Needed some plyers to twist it but easy enough. Straighten it out, outs end in handle, then twist to make the loop. Not perfect but would save you from having to go get nichrome. I actually nichrome but am using it as igniters of my rocket detonator and didn't want to have to go to the store. But when using MSS, I don't bother smearing either. Just put some drops and wrap. Germinates no problem either way.

-jan 3rd .  I checked my plates and there was no noticable growth. One Golden Halo was bacterial. So, I swabbed 3 more Golden Halo plates. I really hope all my plates germinate.
Jan 4th. Checked today and zilch, nada, nothing. Hey that's better than having more bacteria. Turn on, Tune in, and drop out.

Thanks for the tips, guys. I like that you have a visual indicator of spores being on the swab. The print I have is so caked with spores, I had no idea whether or not I was picking up any.
I'll PC some swabs I have with the agar. Woo!

When you guys swipe with a loop does it do anything to the agar?
Idk if it’s cuz my plates are too hard or what but it seems like there’s either no marks at all or the agar splits all the way down to the bottom of the dish.
I poured some softer ones for the morning so I need to learn how to use a loop better.

sometimes it does sometime it doesnt, depends on how hard u press tbh i like stiffer agar personally and dont really like to dig into the agar, keeping the majority of spores on the surface of the plate, again that my way of doing it more than 1 way to skin a cat

when your loop moves across the agar (which appears smooth but at the level of the loop is quite bumpy) the spores are shed from the loop - most don't even land in the direct path where the loop touched the agar - you can see these spores (circled in red) from my streak that are about half a millimeter above where the loop streak is, circled in green:

even if you can't see it yet, they're probably germinating at a turn in the streak where there are clusters of them:

you can see the streak even more clearly here, as well as all the spores in the bend of the path. you can trust that if you at all agitated the surface of a print and then touched the agar or even moved it around near the surface, spores were deposited. You've mentioned a couple times "hard agar," what was your agar recipe?

Ok thanks guys that gives me some hope.
I don’t remember exactly what it was. When I decided to try agar I just got 40 plates and a small bag of premixed lme.
I had made plates a few times and they were always softer than I wanted, so when I made this last round I used the rest of the premix which was slightly less than I had used the time before and then put 5-6 grams of agar agar in with it cuz my bulk agar stuff had showed up.
It’s pretty solid, I’m still experimenting with recipes and this was too much agar. It’s really easy to pick up transfer pieces though, the softer plates I was having trouble keeping the chunk on the blade.
Im thinking somewhere around 5g lme with 7-8agar to 500ml is the zone for me for plates I want grown out ASAP.

Lol pretty much. My first ones were so soft that when I poked the blade in and lifted the transfer piece it would get 1/4 inch off the plate and then kind of slide/melt off.
It was like playing one of those claw machines that always drops the prize.

oh wow yeah that's frustrating! back in the day when it was my job to pour plates we mixed at 20 g agar/L so I think you'll be alright

Holy hell... Haha. I keep seeing other's agar mixes and don't know how mine is as hard as it is. I use 7.5g per 500ml. Loop doesn't dig in at all. I've been thinking about doing a soft batch for nocing up grain. May go down to like 3g or 4g.

Finally got my update in. Hope to get some lab work done.
January 4th, 2020
First signs of growth appeared in at least 1 plate of all varieties on January 3rd. Today, growth is apparent and growing well. APE is I little behind the others but I suppose that is to be expected.
I am trying to make it to my lab now to make the first transfers. Side note: I'm interested to see everyone's transfer plate numbering. That is, whether your spore\grain\tissue plate counts as T1 and first cut T2 or, first cut is T1. I have seen it discussed in here before but I am excited to see it play out in real time. I will only do 1 transfer of each variety tonight, and pour more plates. Then make some more transfer as I go, but keep those first transfer as the lead for this event.
Also have some tamps T2 to transfer (though not for this event) and will start some Z spores as a late addition for this event. I will try to consolidate my log so updates are not so lengthy, though I do tend to blather on about things of little import.

I see some people doing transfers really early. What benefits are there in this. I assume to transfer away from potential bacteria. Maybe my AC could do for one before it gets overgrown?

50g agar holy crap lol, i dont think ive ever done 20g to 500ml... maybe its just the brand of agar I have but I usually do like 10-12g to 500ml and it turns out soft but not to soft..i do 8-9g to 500ml for my germ plates which once again are soft but not soupy soft.

I'm guessing you're using agar flakes then, which are typically instructed to mix at about twice the rate of agar powder
