I appreciate your posts. They're nice.

As far as a drop in the center goes, it works great IF the syringe is clean as fuck but doesn't qualify as "streaking". You're plates are looking good, man.

i have been just adding a few drops from a syringe to a sterile swab and just swab the damn plates, they have enough condensation as it is i dont need shit rolling around on me,
ive used a ghetto homemade loop too, but my swabs come pre-sterilized so thats just the way i do it

I usually just put 4 single drops at the 4 "corners" of the plate. In the process of wrapping the dish they move around a bit. I got sort of satellites at first instead of a smear or streak but seems to work well enough. I use a homemade loop to swipe prints but I see no reason to introduce 1 more vetor into the plates if I don't need to.
Will update shortly with pics of growth.

Loving the microscope shots in some of the grow logs I'm seeing. Making me want to get one myself

Yeah, I should update the terminology. I guess I just see it as a similar mechanism - using the properties of a droplet to skew the present microorganisms by another physical means than a loop.
I would assume (maybe incorrectly) that the bulk of the microbe load is deposited at the point in which the drop hits the media, and then when I tilt it about in a semi-controlled manner to kind of zig-zag the solution around, little satellite cultures have the freedom to germinate unimpeded by that microbe bulk.
Something like 'droplet skewing'.

That.
Of course, adhering to tried and true cell-culturing techniques (like streaking) is a better way to ensure success. But it has worked for me. As verum said, a lot of that likely depends on the integrity of the primary source.

I finally got around to swabbing the other woodlovers that I said I would do, plus an extra. For these I am utilizing some 2-compartment petris, with each compartment used as if it were a full plate. I am using MEA with 1.25% agar agar, after a failed attempt a month ago with 2% agar. The species I added to the party are:

Psilocybe azurescens (3 half-petris swabbed with swab left in fourth compartment)
Psilocybe allenii (3 half-petris swabbed with swab left in fourth compartment)
Psilocybe tasmaniana (7 half-petris swabbed with swab left in 8th)
Psilocybe weraroa (7 half-petris swabbed with swab left in 8th)

Got some stuff done tonight, details here: some single spore transfer attempts

highlight is this lone spore I found and transferred. goal is to workman method it with a big B+ print if it germinates.

I'm going to make another batch of agar and pour again tomorrow. It's totally cool to use a print that you've previously scraped up, right? Also, I'm ditching using the Bobby pin as an inoculation loop and picking up some nicrome wire.

Also I remembered that I ordered a spore syringe sitting around. I happened to have a dish in my batch that I forgot to swab and appears to be clean, so I might try footpaths "drop & tilt" method for fun.

as long as u opened the print in ur SAB it should be fine man, u can use a print many times, just another option if u didnt wanna go the loop route, u can swab ur plates with a sterile qtip, mine come in pre packaged from a medical supply store down the road, but heres a tek on how to make ur own
