Even though I am definitely getting a rough start, lm determined to get the fuckers to grow. I am working with Psilocybe Cubensis Blue Meanie purchased from a vendor in a syringe, which was contaminated with what I am assuming is yeast. They were the only spores I had on Jan 1 because I am really new and I’m waiting for my first monotub to produce my very first flush (KSSS). I’m definitely glad I acquired spores from a variety of sources! The Blue Meanies are going to be a challenge. I also had some good luck with Psilocybe Cyanescens which I am growing mycelium to cultivate in an outdoor bed. I did a spore solution on Agar with them and have done very well. I have some LC of Penis Envy and KSSS that’ll inoculate grain bags and BRF jars tomorrow. I am also working with Blue Oysters, Maitake, and Chanterelles. I’ve been at this for about 6 weeks and these forums are awesome!
Does anyone know what organism this is?

If the solution in the syringe is contaminated with yeast, the yeast grows much faster and produces a goo that the spores either get swallowed up in or float on top of. My only way to extract spores from the goo was peroxide. I will try a drop of the second syringe on my last agar plate and in a few days I can post a picture. If I hadn’t found out about peroxide I would have assumed it was a total loss.

This so cool, well cats! Can copy that picture? And seriously, this noob is really curious how you juggle the magnifying glass and slice out one tiny strand of myc? I'm thinking getting one tiny strand would lead to a guaranteed isolate.

This makes me happy. I used to keep my dog out of my lab for sanitation reasons but he goes EVERYWHERE with me and figured him laying under my chair\legs\table isn't going to stir up enough air to screw up my SAB. Now, he knows the routine and just lays down and watches while I do my work. Love seeing that bound between man and animal in other's lives. Such a special thing to have.

So much cool stuff happening, thank you all for participating in this!
PSA: If you don't have a link to your grow log in your sig, please put one, if possible. I find myself going back to the first post and clicking through from there whenever I read an update that makes me interested in your whole log...

Thanks to CBK, I already feel like a winner. The ESS I’m running are from one of his swabs.
Muda was a stand-up dude and his creation should be shared with the community. It took years for me to get my hands on this gorgeous cube, and I want to share the gift I’ve been given. I’ll be taking swabs and sending them to any participants that want one. Between myself and CBK, we should be able to get this beautiful specimen back in the spotlight

Didn't do any transferring here, but it is still a bit of a juggle getting the right angle of the nearly transparent early germination with a magnifying glass and petri in one hand and camera in the other. I'm usually kneeling, so I use them for some support as to reduce vibrations as much as possible. I have a new tripod on the way though, so this convoluted juggle will no longer be an issue.
As far as transferring tiny early strands to have an isolate, it's probably not very effective except with more advanced tooling. Accurately isolating it from the rest, knowing how well it performs, not introducing something foreign... seems like a gamble when you can much more easily clone from fruits.

Yeah, at least one of the cats are hanging out by my feet when I'm doing SAB work. They're surprisingly considerate of not disturbing me in that circumstance beyond just grazing my legs. They definitely know when it's critical or not - harvest time, they have their heads right in there and bat at cut fruits and whatnot. They wanna be part of the experience, and I'm happy to have them there. I can just about guarantee that they've never been the cause of any errors.

My agar is way too hard would it be worth dripping a drop of sterile water and tilting the plate so it runs down the groove from the swab/loop if there’s no germination in another week?
Also how do I add a link to my grow here in my signature?

Go to your OP. There should be a post # that's a hyperlink you can click on. Click it, then copy your URL.
Above the text box is a button for a hyperlink. Looks like Earth with an infinity sign. Click it, it let's you create the title, then just paste the URL into the space provided. Bam.

I'm sorry you got some infected shit, that sucks! are you inoculating your dishes by just putting a drop of spore syringe on or are you streaking with a loop?

not sure what too hard agar is but i cant imagine myc wouldnt grow on it unless no nutes i wouldn't do that add water thing, sounds like a recipe for disaster.
for link in sig click here: https://www.shroomery.org/forums/editbasic.php?Cat= scroll down to sig edit, then copy your log link from CBK OP https://www.shroomery.org/forums/showflat.php/Number/26388607#26388607
can format that to words you want to see hiding hyperlink by click the little globe+chainlink icon, it will look like this (add open [ and close brackets ] to fix this example)

So, ya, a clone trumps trying to isolate to purity.

So essentially we are isolating to get a contam free strand of MS and then grab some clones off the grow. Correct amundo? LOL
BTW I nominated that awesome cat pic! His eyes are sooo intense. I have a cat with great eye contact, but not many do it.

I would love for my bully, Floki, to hang with me while I work, as would he. The problem is he believes his 80lbs ass is a lap dog and would be grunting and nudging me the whole time to get in the chair with me haha.
MooseMomma, looks like we may be in the same neighborhood, let me know when you get some outdoor patches going, deer have already destroyed a small one I made! Bet they had one hell of a walk through the woods!

Ayy, germination! at least 1 plate from each variety shows the faintest bit of myc. I could transfer out some isolated colonies, but I'd rather wait until I can identify organized growth, and pick an attractive/fast strain rather than guessing. My plates appear clean, so no rush to transfer away.
Nothin really worth photographing yet, ill update with pics when there is enough to see

Just wanted to update to show the efficacy of simply just tilting a drop from a spore syringe as a method of streaking. Plus give big ups to the vendor I can't mention for clean AF syringes.
I fear I'm getting carried away with my posting style... might have to trim the fat somewhere along the line here..
sweet man, aint shit going on in plates
so u just drop in at an angle or u let it kinda just roll around the plate in a controlled manner after u place a drop?
