Cubes: KSSS(FA) KSSS(FA) x RustyWhyte PESA
Looking to stabilize/explore the mutant genetics in KSSS, and hopefully find some rust spored leucistic mutants from the RW X just for fun.
1/1 Swabbed 3 plates with each, for the attempted cross I swabbed the RW print then the KSSS print before spreading them on agar together. (I will do the straight KSSS FA plates first to avoid accidental cross contamination)

Germ on all the RWxKSSS and PESA plates but only one of the KSSS so I’m guessing its mostly RW spores germinating, gonna give a couple more days for mating and to let more ksss spores germinate and then assess mycelium morphology before making any transfers. Will prep dishes today so theyre ready and waiting.

Preparing to take 1-2 transfers from each t1 plate, most will probably be ready for grain on the next plate.

Most excited about this one, havent seen any RW myc that looks like this before, and its exactly what the myc from the ksss clone the spores came from look like. Hoping thats a sign I got a cross, but either way its got some nice clean lookin growth.

These Pes-A sure have some nice rhizo’ sectors, Looks similar to some of the aggressive colonizers I’ve grown before. I know where I’m transferring from with these fasho

The straight Ksss spores still have only germed on 1-3 plates, and only three small spots of germ, kinda makes me wonder if any are monokaryons, and how many actually germinated on the mixed plates? But I’ve heard other mycelium can help spores to germinate, so I’m hoping thats the case on at least some of the RWSS plates. Should ne easy enough to tell once they fruit. Until then I might just be cleaning up RW myc.

Just dropped all these to grain

Making transfers on these tomorrow. 1 more and theyre most if not all ready.

1/30 Progress from plates dropped 5 days ago, will be shaking in 2-3 days

last round of agar, all getting dropped to grain to sink or swim also in a few more days

That pesa looked so good I couldnt resist doing some g2g’s yesterday with jars @ 1:10 and bags @ 1:3


And this pesa plate pinning fast AF got me excited

The RWSS from t2 didnt look as nice as the pesa but I’m still gonna spawn em and see what happens

Shaking everything from t3 plates RN (Pre-shake)

Things got busy for a while but back on track now!

Finally got an update with fruits

Also took a pin clone from a Different PesA plate and its already fruiting again!? This thing is fast AF, dropping a plate to grain today, I'm guessing 10-12 days from spawn to fruiting, usually takes 2x as long as this to pin on agar and I’ve never seen a second flush on agar like this wtff

Rw x ksss pin???

Maybe just RW but thats a thicccy stipe. Worth cloning? Probably just gonna wait and see what comes in the ms

Still only 9:40 over here on the best coast, sliding in with time to spare

You can use it straight up but I like to dilute it for clarity and on weaker agar myc tends to run a little faster.
Dont think anyone has gone through the effort to do the experiment to see which performs best. But since everyone probably uses a slightly different amount if water to grain when prepping IDK how accurate it would really be anyways. I just noticed if you water it down about 1/2-2/3 the myc starts to run faster which means its seaching for nutes and this helps for cleanup transfers but if thats your only goal you can make agar with straight h2o and use that for a transfer. Theres enough micronutrients in the agar itself the myc doesnt die but it does spread fast looking for more food.
There is no “right way” or consensus. Depends on your prep, the grain, and your intended use for the water. You can water it down even more for LC and it will still work decently. 100% gwlc works too but it takes a while to colonize.
Sometimes you just gotta figure out the ballpark and experiment from there to figure out how it will work best for you

Yay spores!!! Lol. Thats the whole cultivating part tho right? Selecting for certain organisms to succeed over others, people often misuse the word to simply mean growing a thing without realizing the connotation, that cultivate basically means to remove competition. Its actually a pretty aggressive term that is often used in a very passive way.

If you wipe the blade off before flaming you wont have soot. Its just carbonized/burnt agar. Doesnt rwlly change anything and its sterile so I usually dont bother

Unicorn bags all the way for me. Out-grow (sponsor) sells them individually, but theyre cheapest direct from manufacturer, also a member here named Gr0wer has a website called myers mushrooms and he sells them by the 100 with a decent price.
If you can swing it and use enough of them, its cheapest to order direct from unicorn, ~400$ for 1000 bags. And if you’re an e tra cheap skate they sell factory seconds for like 80$ for 1000, the website says they will only sell them 10 boxes at a time but call and ask, they’ll ship you a single one. About 100$ after shipping and 9/10 of the bags are fine but definitely inspect each on thoroughly.

Cook oats, make sure theyre dry on outside, load bags, accordion fold or preseal and PC 2.5-3 hours at 15 psi. Bottom layer folds up rest of the layers folds down. Make sure your pressure doesnt drop too fast or bags can pop.
Pretty much the same as making jars just a little bigger and easier to damage.
LAGM2020 update: Poured agar for my t2’s last night, will do those today and update.

Sadly we'll never know - I dumped it right after transfer. Could definitely have been mold, but it looked just like newly germinated cube myc (somewhere between wispy and rhizo) and where it intersected the leading edge of the PE myc neither seemed to care at all (which made me think it might be the same species). Trich is a parasite and will gladly grow right alongside and intertwined with it. Alot of what people call trich on this site is actually aspergillum and penicillium, neither of which are great but IME they dont spread to anything healthy and mostly grow on rotten fruits or grains that werent fully colonized.

Re-doing everything today. Not fun.
Try ramping down the heat or wrapping the presto in a blanket after you cut heat. The popping is from rapid cool off. I also used to tie my bags up in a pillowcase when I was using presto’s.
To be completely honest I used to lose 1/6 bags pretty much every presto run, I’ve yet to lose a bag in my AA941, but I’ve popped a couple in my 921. Always happens on cool down because pressure difference between air in cooker and air in bags. If they stretch but dont pop I’ll flattencit out and bandage it with MP tape before inoculating, so I dont accidently stick my finger through it or tear it while shaking.

You should try taking smaller pieces of agar, the size of a grain of rice or smaller if possible and use one piece per new plate in middle of the plate. And that transfer looks dangerously close to that mold colony, he suggested 6 o clock and it looks like you went closer to 4

More Fae That shizz is everywhere all the time it only thrives in wet stale conditions. No need to shut down just put a fan in the room. It isnt airborne but can become Airborne in water droplets if you mist it but it wont go far. I see it in alot of people's grows. Its only really an issue if conditions are shitty. Also totally safe to eat just a little unsightly, you could also try prepping your sub with a bit less h2o
I think of it kinda like bud rot (botrytis) when growing weed, you dont want to see much of it, but a little just means your getting nice super dense buds, or in this case a little pseudo because your clusters were so dense.
