2 • 13 • 20• Spores → Agar • Here we fucking go again - new source for Chitwan sporeseses.
Some sloppy technique in this video: - 0:47, I start monkey-gripping the syringe - hovering my hand over the sterilized needle, nullifying the sterilization. - 1:11, I have to pass off the swab from left to right hand - not a huge deal, but I should familiarize myself with doing it opposite (either streaking or dropping spores left-handed) 2 • 18 • 20• Germination Day 5 • Now it's looking like a normal germ plate. I'll let the colony at 2 o'clock grow out a little bit and take it from there.2 • 22 • 20• Germination Day 9 - Transfer 1 •
Surprise! It grew out more! That 5-6 o'clock range in that 2:30 colony looks best. Probably should have grabbed it a bit earlier. Fucked up a little by not taking all the way into the leading edge, and not quite into the 5 o'clock range of it. Whoops. 2 • 25 • 20• T1 Day 3 • Growing out alright - decent vigor, predominately nice looking. I'll need to make up for my botched grab at that germ plate with another transfer or so. Nevertheless, It's nice to know I've got something familiar.3 • 2 • 20• T1 Day 9 • Steady as she goes. I'm happy for them being T1's. Two of three look pretty enough for me to expand. I'll wait a couple days and take T2's, make some LC, and transfer some to grain.3 • 5 • 20• T1 Day 12 • • T1 → T2 •• Agar → Grain •• Liquid Culture • Eventful day for this culture. Might be diving in a little too deep for a first transfer but... That's why I took T2's.

Maybe I'm out of turn, but I think the most important takeaway is the communal aspect of it all.
It could be a place for some to light a fire under their ass to get started on something new. It could be a place for novices to constructively compare notes with more seasoned growers.
Ultimately, it should be a place to bring us all on the same damn level - we're all just growing shit, and that shit's fun. And we should all be happy to see each other growing that shit. Whether you PF Tek Golden Teachers or do supplemented sawdust logs of an exotic woodlover, we should all be able to encourage each other's growth. And I really appreciate that CBK has set up a platform for us all to do this on a relative timeframe. It better helps in the learning process and in the fostering of community.

It'll be a good show for the thread. ...that sounds really sarcastic.
But for real, just roll with it. People, especially the inexperienced, need to see how agar should be used so maybe we see a little less of the antibiotic agar threads.

I have literally one drop in this. Which dropped ~7PM 1/1. So I guess I can still have germination within two days. But I don't really recall ever getting that growth rate with a syringe.

I'm not of the competitive variety, so if I happen to 'win', please offer it to the next in line. Those ESS look do look pretty though...

Inthepit said: This so cool, well cats! Can copy that picture? And seriously, this noob is really curious how you juggle the magnifying glass and slice out one tiny strand of myc? I'm thinking getting one tiny strand would lead to a guaranteed isolate.
Didn't do any transferring here, but it is still a bit of a juggle getting the right angle of the nearly transparent early germination with a magnifying glass and petri in one hand and camera in the other. I'm usually kneeling, so I use them for some support as to reduce vibrations as much as possible. I have a new tripod on the way though, so this convoluted juggle will no longer be an issue.
As far as transferring tiny early strands to have an isolate, it's probably not very effective except with more advanced tooling. Accurately isolating it from the rest, knowing how well it performs, not introducing something foreign... seems like a gamble when you can much more easily clone from fruits.

I used to keep my dog out of my lab for sanitation reasons but he goes EVERYWHERE with me and figured him laying under my chair\legs\table isn't going to stir up enough air to screw up my SAB. Now, he knows the routine and just lays down and watches while I do my work. Love seeing that bound between man and animal in other's lives. Such a special thing to have.
Yeah, at least one of the cats are hanging out by my feet when I'm doing SAB work. They're surprisingly considerate of not disturbing me in that circumstance beyond just grazing my legs. They definitely know when it's critical or not - harvest time, they have their heads right in there and bat at cut fruits and whatnot. They wanna be part of the experience, and I'm happy to have them there. I can just about guarantee that they've never been the cause of any errors.

verum subsequentis said: As far as a drop in the center goes, it works great IF the syringe is clean as fuck but doesn't qualify as "streaking".
Yeah, I should update the terminology. I guess I just see it as a similar mechanism - using the properties of a droplet to skew the present microorganisms by another physical means than a loop. I would assume (maybe incorrectly) that the bulk of the microbe load is deposited at the point in which the drop hits the media, and then when I tilt it about in a semi-controlled manner to kind of zig-zag the solution around, little satellite cultures have the freedom to germinate unimpeded by that microbe bulk. Something like 'droplet skewing'.Quote:
tealseal said: or u let it kinda just roll around the plate in a controlled manner after u place a drop?
That. Of course, adhering to tried and true cell-culturing techniques (like streaking) is a better way to ensure success. But it has worked for me. As verum said, a lot of that likely depends on the integrity of the primary source.

Most contaminating shit isn't going to have the kind of edges that carbon deposits have anyway. So, aside from aesthetics, it's really no issue. I guess, worse case scenario, something could be hiding under one of those flakes.
Alternately, you could use a cooling plate:

I like to see how long I can get away with using one until it contaminates, lets me know my technique is on point.

All good. Assuming it's a gas range - it's either propane or natural gas... same as bunsen burners or torches.

Grocery store cloned Blue Oyster:

Even the first transfer away from the original clone was totally clean. Well, either totally clean or the BO destroyed everything else in there. Also fully colonized probably within a week. Oyster is crazy aggressive. Too bad I haven't put these to straw yet... just a bunch of petri fruits and waaaaay overripe spawn jars.
