Any way to take and preserve clone culture without agar?
    #26385727 - 12/16/19 06:48 PM (4 years, 2 months ago)

So I have a beautiful mushroom I would really like to clone, but I won't have my presto 23 until christmas. (It's a gift) I'm trying to find out if anyone can think of a way I could clone that mushroom without being able to make agar plates. It still has it's veil intact so I may have a day or two.

What if I were to boil a piece of cardboard, put it in a jar with a tyvek vent, steam sterilize the jar (like the pf tek), and then take an inner stipe sample and put it in the cardboard.

Sort of the same principle as the cardboard spawn tek but using the method to create a clone.

Anyone think this would work?

Anyone have any other ideas?

Do you have agar? I'm sure you can boil it for some length IDK exactly. You can also make a flour puck, BRF or otherwise.

Yeah do a small amount of agar boiled for longer than usual. If no agar then make an lC, hell even straight water should keep it fine till then.

Even if what you do is dirty, clean it on agar later when you are setup.

Try all recommended ideas so as not to have all your eggs in one basket. Also you can take a clean sample and place in an unused zip lock bag to transfer at a later date. I've done this successfully when cloning wild species to test if samples revert to vegetative growth before placing on agar.

I have agar and lme I just don't have a way to do no pours. I also don't have petris or sterile containers to make into dishes.

Thanks for the ideas guys I will try them!

Christmas is right around the corner. Throw that fucker in a paper bag and toss it in the fridge. It will keep fine until Santa hooks you up :super:

Buy the same Presto 23 on Amazon.  Use it to do your thing, then when you get the gift one, return it and pretend the first one you got was the gift one.

Hahaha you guys are awesome.

Can I really just dry the mushroom then clone it after christmas? I was talking to someone on reddit about this and they said that almost nobody has successfully cloned a dried fruit and that it isn't a reliable route. They very well may have been full of shit.

Also I didn't know that it will stay fresh enough for up to 7 days in the fridge in a paper bag. My fridge is kind of nasty lol.

Awesome Cronicr!

I think I may try both by cutting a hunk of the stipe off and drying it while putting the rest of the intact fruit in the fridge. Then I will put both on agar for practice.

One question if you don't mind my asking:

How long did you soak the dried one? Your thread said longer than the fresh ones but I didn't see a time estimate.
