Analyze my Agar?

Was hoping someone could take a look at these plates I made up. All of these were made scraping spores off the foil in front of a flow hood and on top of the agar. Some excess spores that built up on the tip of scalpel were dragged through agar on some, but for the most part just sprinkled the spores on top.

The first Fiji, and PE6 I'm pretty sure are good, the GT (also the only print I bought, from a sponsor, the rest were from Shroomery members) shows pretty awesome growth and feel the most confident moving forward with. I'm not entirely sure what happened with the 2nd Fiji, but the slime around the drag marks appeared within 24 hours.

The RW obviously has something wrong with it, and this is my 2nd attempt at RW spores to agar and both times it was prevalent contaminates on the agar :frown: The Huatla plate I'm not sure about, when I looked at it about 2 days ago it didn't have the coloration in the middle, but looked questionable. Now today, the leading edge looks "good", but I'm not certain that it's mycelium.

I had one more plate that I left out which was a print called Armageddon, but nothing germinated. It was also the heaviest spore print I had and it absolutely covered the agar with spores.

take some transfers from clean looking spots.

For the rw, you might try using a swab on the spores (instead of scraping) and make a streak so you have a better chance at separating the good from the bad.

What are you planning on doing with these?

Yes I have a flow hood. This was my first time working in front of it actually.

I left three plates open in front of the hood to test while I put the spores to the rest of the plates, and none of them have any sign of contamination in them a week later :thumbup:

I plan to do at least 4 generations of transfers with each of the plates that I am able to get a clean sample from. From there I will probably do agar to grain with the best plates, and more than likely make a rack of master slants to preserve them for long term storage. I was maybe considering doing a final transfer to LC, but I am not a fan of using LC after my last few attempts. I think you can get more consistently with strictly agar to grains rather than LC to grains in terms of assurance of sterility.

Also not sure if I will make slants with the final transfers, or if it would be a better idea to make a bunch of trays with my final transfers, and then clone and isolate from fruits, as I do know there's potential to isolate shitty genetics so it may not be worth making slants until I know I have a good performing culture.

On the subject of your plates, things look pretty good. In the future, it would be easier to transfer clean growth if you streak the plate as opposed to sprinkling the spores on it. It becomes harder to see things when there’s so much going on. If you want a clean RW print, PM me your addy and I’ll get you hooked up.
