Haven't tried cheesecloth or paper towel, but the coffee filter cannot absord a lot of the liquid, it is saturated after absorbing some mL's of liquid. I usually rinse it with water first as well. I've never worked with LME though so I don't know the deal, however I feel like I've read on here that LME should be clearer than the alternatives.

Thats a pour with LME I did yesterday and its clear with no filtering/straining. Thats about 12ml with 10g Ag:10g LME:500ml water.

Maybe you do just have a bad batch of LME? I also mix my LME with a bit of water into a paste before I pour the rest of the water in so it doesnt clump, not sure if that will help at all.

Yeah, I bought both my agar and LME (btw, I mean Light dry Malt Extract not Liquid Malt Extract) at some random cooking supply store and they aren't great.
I got it super clear once but it took a lot of filtering and decanting.

Zakkery, do you boil the LME before pouring to the HG?

That's great to hear! I'll have to give it a shot myself. How soon after PCing them do you move them to the dehydrator? Did you prep the dehydrator in a special way first?




I think it was that same day that I loaded them in the dehydrator.  It took a while running it at lower and lower temperatures to ensure the condensation didn't come back.  I didn't clean the dehydrator out at all.  The lids weren't really loose; only slightly backed off from tight.  It was a few weeks ago now and I still haven't put them to grain yet.  They still look clean.

Really surprising. Lids loosened, and blasted with warm air? My dehydrators are filled with all kind of spores lol.
How long did you monitor your plates before deciding they're clean?




It's been a few weeks and still no contams.  The lids were slightly loose, not really loose.  I didn't clean out the dehydrator first.  I was actually very surprised too that there was no contamination of any of them.  All the plates were from the same cloned cubes.  I kind of doubt that the ones I put in the deydrator ended up as multispore plates due to the residual spores that were in the dehydrator from drying.  I am putting some to grain in the next day or so and giving the others to a friend so he can do the same.  I will make sure to keep track of which jars have the original condensation-free agar and which ones were from Odo-dried plates and see if any of the jars contaminate.

I have dry LME too. First I mix the LME into a paste with about 100ml water, then add the rest of the water and some colouring, followed by the agar (all in a jug). Then I pour the jug contents into a pan and heat it to a simmer to get it all mixed together nicely. I normally kill the heat once a thin foam starts to form on top of the liquid. Then I use a little 12ml scoop to pour the plates.

I just did a batch earlier today, I'll definitely try your way next time.
I think I'm boiling the agar/LME too much, your way seems better.
This time I noticed just how murky the water gets when I mix in the LME, I was thinking of mixing the LME in water and filtering in advance and keeping it in the fridge, then just add to agar.

Okay, here comes a long post with some conclusions. I will go ahead with the tldr right up front.
TLDR; the PVC tube helped, the capped PVC helped more, stick to alien’s instructions about pulling them out hot.

So as I mentioned in a previous post, I found 3 inch diameter by 2 foot lengths of schedule 40 PVC at Home Depot. Schedule 40 is important because it has a higher melting temperature. I found that my 23 qt Presto with trivets placed to keep everything above the 3 quarts of water allowed me to stack 9 holy grail plates nicely in a 9 ½ inch tube. So out comes the saw. My first test attempt was with a cheap hand miter which was too small and failed to give me a straight cut. But that was good enough to verify that they tube would hold up to pressure cooker sterilization without melting.

Thankfully, my son reminded me that I have a circular saw miter thingy which made quick work of the rest. Nice straight lines.


On to the first agar test. I wanted to compare the results with and without a cap on the tubes. So I bought one 3 inch pvc cap. I don’t think it is actually schedule 40, but I took a chance that it wouldn’t melt all over the hg plates. It looks like capped or uncapped, 7 tubes will fit in my cooker. Also note that the capped tube top meant that the weight of the tube rested on the top of the stack of 9 plates.

I hoped that would help by applying a bit of pressure on the top which seemed to make small bit of difference on some of the plates in jars for others. Alien suggested capping both ends but I was a little concerned about the pressure build up in the double capped PVC. In the end, I could not remove the cap from the post sterilized tube, so I am glad that I did not try that yet.

I also have some pyrex dishes that I had never used, so I tried some no pour with them in the same PC run. So I poured my holy grails and pyrex dishes with unsterilized agar. Wrapped my pyrex in saran wrap per Poisoned’s no pour tek. And put it all in the PC for 30 minutes at 17 psi.


In my infinite wisdom, I decided that letting everything cool overnight in the PC would be fine. It was not. Out of two set of 9 uncapped, 6 of each had lots of condensation and/or puddling vs 3 that were okay. The capped tube was marginally better with 5 wet dishes to 4 pretty good and clear hg plates. The Pyrex gave me 1 or 2 good plates in each stack of 6.


That is why it took a while to get this report back. Time for test 2. This time I followed Alien’s instructions and removed the hg stacks after the PC pressure had dropped but while it was still hot. I gave it an hour after the knobby thing fell indicating 0 pressure before opening the lid and removing the tubes. My infrared thermometer showed that the outside of the tubes were around 155 f and the two agar media bottles that I decided to share the PC were at 195 f. For giggles I placed the hot media jars on the top of the two tubes to cool overnight. That was a bit of a cheat, but it probably got me an extra clear plate in the morning.

So the body count on method two was better. 5 out of 9 look really good from the uncapped tube.  And 6 out of 9 are also nice from the capped tube. The ones with condensation still look far better than the wet messes from the first round that cooled in the PC.


I guess the next thing to try would be the double capped tube. We just have to solve three problems that I can think of. 1) venting holes – are they needed and where 2) how to get the effing cap off of one end after they have been PCed 3)may have to reduce the number to 8 per tube to accommodate the additional height of the second cap.

Hello all. I'm definitely having no luck finding these HG containers at the moment. They look awesome though. To be honest, the Ziploc Twist 'n Loc minis are working very well for me these days. I do a side hole with two pieces of micro pore tape and use no foil or any sort of covering. These could probably be used unmodified as well, similar to the POD tek which I have goofed around with successfully. I have also used the pint and quart sizes as unmodified grain spawn containers. Just barely loosen the lid before PC'ing.

For these minis, I do use two PC racks and elevate the first layer of them out of the water a bit with a few shot glasses though. These aren't as cool as the HG appear to be, but they are pretty great nonetheless. They stack well, require no foil, and you can fit more in the PC than the glad minis(no awkward lip). I have condensation, but it honestly is not a big deal. I get away with quickly dumping it out in my SAB without any contams so far, and if it is a germination plate or plates, I just very carefully nuke them at 5 to 10 second intervals until the agar liquefies(be very careful to avoid a boil over though). This clears up 90% - 100% of the pooled water.

That reminds me, I actually meant to nuke a stack of HGs and see if I can clear up condensation/pooling by remelting the agar.
I did it to one HG, it boiled over and I was too pissed to try with another one after cleaning the mess.

So i made a whoopsie doodles that resulted in liquid agar touching the lids and leaving a film around the top of the container .... Does anyone find these leads to contams?

This looks absolutely incredible! I wish I had found this before I bought expensive glass petri dishes & media containers...

For using these do you screw the lid on completely through the whole PC cycle & once you knock them up? Or should I slightly unscrew them in the pressure cooker & rescrew once done? - I.e. what's the best way to manage gas exchange screw-wise? :tardsex:

Stoked to be able to use something that is easy & doesn't make a shit ton more plastic waste. Thanks for the write up!!
