Grains not doin so good
    #26373631 - 12/10/19 05:19 PM (4 years, 1 month ago)

Hey Everybody

I’ve got these jars inoculated with small agar chunks on 11-13. They just now look about 100% colonized. They look so wet in there though I’m kinda grossed out, not even sure if it’s worth spawning... this is my first attempt at grains since pulling all my cultures out of cold storage. My technique may be a little rusty.

Golden Teacher 12/10

APE 12/10

Golden Teacher 12/1

APE 12/1

In the past year I’ve had much better colonization times and made it from petri dish to fruiting in shoeboxes with a high rate of success. Starting 7 jars at a time typically, I went 21/21 with no contaminations until after the 1st flush... not sure what happened here but all 7 of my jars inoculated on 11/13 look sketchy.

Any input is appreciated

Those don't look bad to me.  It looks like they haven't finished colonizing is all

Maybe a tad too wet, but not bad at all.  Id give em one more good shake.

It's only been 9 days. You're on par for agar wedges.

The first jar looks iffy.

A lot of my jars that have water being excreted out seem to be contaminated. They start out normal then the water pooling begins..

All those pictured were inoculated on 11-13, that’s 27 days

I’ve had full colonization in 14-21 days with larger and/or multiple agar wedges in quart jars

I’m surprised you think the first 2 pictures (taken today, 12/10) still look decent. I just shook one of them moments ago. They just look incredibly wet to me, somewhat slimy but the liquid is not overly viscous. Maybe it’s just me, I’ve only done grains a handful of times so I’m figuring out what normal is.

Here’s the Golden Teacher jar, just now after shaking

Thanks for the input!

Sorry I misunderstood.  The dates above the pics I assumed were the day you inoculated.  Didn't;t read carefully enough.

They have a decent amount of bacteria.  If its all you got, can still spawn them and see what happens.  Just don't expect too much.

A little Condensation is pretty normal especially when the grain is overcolonized.

As stated earlier, looks like a little bacteria also...spawn it anyway

Ok thanks so much! I probably would have tossed to be on the safe side, but I did just prepare a bunch of coir that’s cooling down in an insulated cooler right now. I’ll be mixing them up tomorrow, I’ll post results later

Is this perhaps a symptom of grains not being dried enough after boiling? Too much moisture in the jars to start with? I find it unusual that all 7 of my jars, all with 7 separate cultures, developed the same problems. That makes me think something went wrong with the grain prep. I follow BOD’s oat tek. I live in a moist climate and it seams difficult to get my grains to air dry. I  spread them evenly on a large window screen and  setup a fan to help evaporate moisture...

Boiled too long maybe?

Probably had to do with your grain prep.  Oats have to be completely dry before PCing.  If rye or wheat is available in your area for a good price, I recommend them because the prep is easier and more forgiving.

If you processed your jsrs properly the extra moisture doesn't cause contams, but rather provides ideal conditions for contams to thrive. If you've got contams its because you introduced them somewhere along the way. Did you take transfers from your cold plates or just throw it right in?

The first ape looks like it’ll do well but the other ones are super sketchy. Worth a shot in a shoebox or prob topfruit for the worst ones.

I’m in the middle of spawning to coir in shoeboxes now. 2 out of 7 done now, 5 out of 7 still in jars.

Do they look fully colonized?! There’s a few spots where the grains are pressed against the glass that haven’t fully been enveloped by the mycelium. This seams to be a regular occurrence in jars though, I’ve seen the same issue multiple times.... Good to spawn today?

Should I shake and wait another day or two before I spawn the rest of these jars?

They smell like normal happy mycelium, that was a pleasant surprise

They’re def bacterial but you’ll prob get at least one flush out of them.

I wouldn’t shake them it’ll just spread the bad stuff around. The reason there’s those uncolonized spots and they look wet is cuz that’s where the bacteria is.

Def use a low spawn to sub ratio so it colonizes quickly

Do you think they’re ready to spawn today, or should I wait awhile longer?

Yeah I’d say go for it on the first two maybe wait a couple days on the second two.

I transferred 1-2 times after removing from the fridge (about 5 months in storage). All my cultures are clones (F1) from my best performing shoeboxes

Those two I would spawn now I don’t think they’ll get any better.
