Isolation Question

So I lets say I have 2 Multispore plates. And I transfer 2 tissues samples from each plate demonstrated in the pics below .






Im wondering if there is any specific reason that I would want to label the quantity of passes from each plate as i go .

For Example , Instead of just haveing x2 plates labelled B+1125(L)P1
(1125 = date, P1 = pass 1 or transfer 1) Should i label them, B+1125(L)P1(a) & B+1125(L)P1(b). The a and b letters representing that there were 2 transfers from 1 single plate?

I feel like this will get hard to continue on Because the next plate would have to be something like, B+(date)(L)P2(a)-1? And then after that B+(date)(L)P3(a)-1-a and so on creating a very long code.

If you take 2 transfers off 1 plate , should those 2 samples be seperated by labelling?

Hopefully I was clear enough.

I keep an (encrypted) electronic journal where I enter the new plates and give them an ID. This ID is then written on the dishes. This has two main advantages:

- IDs are much smaller than date + species + transfer +.... so you see your mycelium better
- If you change your naming scheme, you just need to change things in your journal, the plates keep their IDs

WHy is it important to be able to trace back to exact plates once you have isolated to a "pure strain" ?

Isnt that "pure strain" what you want? not all the old plates leading to it?

Let's say you have a contamination after 3 transfers and you realize also transfer 2 is contaminated. Then you can go back to #1.

Also, with every isolation it's not uncommon to "lose" some "good" genetics. If you can't find a good fruiter, you might want to go back to the first ancestor and start over again.

I'm sure, others might have additional advantages

On the other hand, it's up to you how big of a record you want to keep.

Totally depends on your goals and how detailed you want your "culture-journal" to be.

There is nothing you "should" do. Only you can decide what you want to do...

Ahh i see. So its important to trace exact lineages . Iv been stressing over this a little because it starts to get really confusing.  I guess my goal is to have as perfect records as possible so I can know the origin of anything I want to. . Thanks for the help man.

So every single plate has a different ID number starting at "001" for multipore plate?

"parent I.D." Would be the last plate that the transfer came from ?

Whatever is descriptive to you or leave it blank after the first plate. The original plate will be traceable and "transfer" implies A2A anyway.

Lots of companies have entire departments partially dedicated to tracking this kind of information. If it seems a bit complicated even at our "low" level, that's because it easily can be.

I’m running into this same issue very soon after starting agar, a couple plates deep and 1/3 of the rim is ID stuff already.

The computer/journal is for sure the way to go.

i made a codex thats something like this.
on the plate it will be labelled.        " B+(A)P1-001"
that's as long as the code will ever get .
(A) represents the ms " mother " plate since I usually start more than 1 . p1 is transfer and 001 will guide .e to my codex on computer with dates and notes

The spore plates just get a name and a date no sense writing t0

If i make five transfers from a spore plate i introduce letters

T-1b
And so on.
If you wanted to branch from there you could do

aa
ab
ac
..
ba
bb
bc

It really need not be something you think too much about keep it simple and easy
