Storage of inoculated petri dishes
    #26365112 - 12/06/19 01:43 PM (4 years, 1 month ago)

Ok so today I scraped 4 spore prints onto 4 Petri dishes filled with yeast hull malt extract agar blended medium. I've been keeping the dishes in the fridge since I poured tem a week ago.

Im not sure how to store these dishes for colonization.

Should I keep them in the refrigerator til they colonize or on the shelf at ambient temperature? My house is about 72 degrees farenheit on average.

Also Im going to do a tissue culture in another dish same blend. Same question regarding storage for colonization?

Room temp. Fridge will severely slow growth. Use cold storage for good cultures you want to save for long periods of time.

Put them on the shelf for sure. You only want to put them in the fridge for longer term storage because it slows growth

laying out on shelf room temp always been good to me.

when you store in fridge, if not doing a master slant.. i would wrap them tightly in plastic so they don't dry out fas, also they will be less likely to contaminate protected against the air current in fridge.  a petri wrapped in a couple black garbage bags should be safe for a few months..  anything longer you need to master slant it.  and never save rye or other bulk innoculant in fridge for long, they contimate easily.

Thanks for the feedback yall. I forgit to Mention that the dishes had so e condensation on the underside of the lid from the cooling period. Anyone know if that will be a problem or issue of any kind?

Or put something warm on the lid for a minute. Clears it right up.
