Yeah... again, this is more convenience for the grower than necessity for colonization... not an issue with pp5, but quick heat/cool cycles are hell on glass jars... I usually pull the following morning, but wait a minimum of 4 hours before unloading in flow to shake and let finish cooling before inoculation... td he jars are closed inside the workstation for this, but probably not necessary

First run left me with very shakable pints. Im setting up to run the other halfdozen empty pint jars I have along with 2 quart jars with double the amounts. Got a half dozen SAGARI Pastys growning out. If all goes well this will be my go to method. Got 10lbs of rye berries sitting here with no ambition to mess withthem if I dont need to. WBS PPM seems to be much eaisier.

The further away I can get from pumping syringes into cakes the better. Having a terrible time getting them to grow out on agar. Luckily the LC I made from my first syringe took to a plate and managed to get to T3. One set of genetics to work with. 3 shoeboxes spawned from brf and MS and 1 from that LC on popcorn. Made alot of mistakes in the beginning. Ill be happy if i get a few fruits and prints and clones from both MS. With PPM and agar I should be able to fine tune a more reliable process and keep the syringes in the drawer until I feel up to starting all over from brf. Honestly my collection is all pretty basic. No PE or albino so I dont have alot of desire to secure 5 sets of similar varieties. Ill be happy to hone my skills and focus on technique and yieldsuntil Im in a position to trade or purchase some rust whyte or a PE variety to play with. Thank you Caps for making this process less painful.

I hate to be the bearer of bad news but T3 is a LONG way from a single set of genes: even if you start with a clone... Even a tiny tissue sample from a single fruit will contain hundreds, likely THOUSANDS of individual strains (sets of genes) a true isolate is MUCH more work than it's worth IMHO... Concentrate on a CLEAN culture, then fruit that, pick the fastest and largest from a cluster to clone, and transfer from the tissue sample a few times before fruiting for testing... I suggest 3-5 clones just to save yourself time on a bad pick...

THX for trusting your mushpuppies to my processes and I lol!

The quart jars seem to have come out just as good as the pints. Got a big plate of oysters almost ready for grains. Might go ahead and split it between the 2 and play with that while I clean up my cube plates. i'll figure out how to post pics so we can watch them colonize.

Two of my bags using this method:

:wow::jawdrop:
:amusedapplause:

Have been using boiling water and allowing them to sit for an hour, then giving them a shake before cooking: really seems to have reduced cracking jars... This is amazing and inspiring work BS... Been wondering about quarts and bags myself... The pints do clump but colonize just fine... Really not as big an issue for the culture as it is the cultist... It is more difficult to ensure cleanliness, but when you GANGSTA!? :shrug:

Eh, turns out I'm not so gangsta :frown:. I spawned those 2 bags, both contam'ed in tubs. I've been trying to juggle 5 strains since I started around Feb '19. The only ones I've had luck with are B+ and GT. The 3 other strains have had constant contam issues or just weak. I'm tossing all my other cultures and focusing on just those two, and probably going down to just one.

I'm also going to try a 24 hour soak instead of the prepless bags, just for kicks.

Re: your folding question, I've tried both folding the bag with a tyvek envelope, and sealing the bag. I settled on just folding. I can just pull out the envelope to open it. I like not having to mess with scissors when I open it to nocc.

You still gangsta... pushing the envelope of science here! What's that thing my mom used to say if at first I don't succeed?
