How thick do y'all pour?

About to pour tonight and I want to know what is the best depth of agar?
I poured some pp5s and WAY over did it. Making transfers was such a bitch and it took over a week to colonize the wedge before it started to colonize the new plate... definitely wanna avoid that again!

Not very much. Enough so the bottom is covered, then a tad more.
I think I average about 40ml per 10cm plate. About 1/4" thick

I like to pour about 3-4mm thick. Any less makes it very hard to transfer the wedge safely to a new plate. Protip: Flip the wedge when transferring so the mycelium is directly in contact with the agar of the new plate. No need waiting for colonization of the wedge.

Ya that sounds good in theory but its hard to manipulate the wedge using only a scaple... do you use something else?

It comes quite naturally; I pierce the wedge from the side, take it to the new plate, turn the handle of the scalpel 180degrees and push the wedge on the new agar. Pull back the scalpel and you're done.

Ya I usually poke it from the top and sit there and shake/tap it until it falls off...total bitch.
Or I use a tweezers but that's just one more tool to keep sterile...

yeah I shoot for 10-20 to make it stretch. you can go down to a 15 g/l agar mixture and it's easier to pour a nice thin layer

1/16" normally - 1/4" if you want it to last long without drying. I find about 1/16" is perfect most of the time. Thinner can be trickier for pulling transfer wedges when you're new at it however.

I pour mine about 1/4" in my jars/pre-pours. In a petri... maybe 1/16" less or even as thin as 1/8" ... I use my petris for initial transfers/cleaning up however, and my jars for longer-term storage in the fridge.

Yeah wtf is with all the no-pour teks talkin bout “pour as thin as possible”? I had to use my scalpel and inoculation loop at the same time to make a transfer from my first 1-3mm thin plates lol. It looked like moldy skin👌 Made it too soft as well though..

i use only pasty plates and in the last 2 months i started pouring them real thiccc
i've gotten way better results after doing this. Makes transfers very easy. Before when it was thin, transfers were a pain in the ass due to the height of the pasty plates.
I don't notice any problems with colonization or anything - may need to dial in your recipe
Sometimes when taking transfers I will just skim a little bit off the top instead of taking a full wedge

Right? I like thicker for that reason as well in my jars because I use those little standard 4oz(?) jars (not the squat, expensive ones)- and when I take a wedge, the angle of entry isn't convenient for fishing around for a thin one. I like a reliable stab-and-done (so much easier a little thicker- for me anyway). I know, I know... my technique isn't terribly seasoned... but I can deal much better with a thin wedge with a petri for sure becasue I can slightly "lift" the very edge of the wedge at a low angle with my exacto without cutting into the side of the wedge before stabbing and grabbing to avoid it just sticking in place when I stab. I need to just buy a case of petris... I make mine fairly firm too btw.
