Psilocybe Promiscuity * 6

I just thought I'd briefly share my experience using different grains for Psilocybe cubensis. This species is particularly promiscuous in that it will grow on almost anything I feed it. For this particular experiment, I did a side by side with hard red wheat berry, barley, popcorn, hulled millet, and a mixture of all four (left to right in the images below). The mycelia were more than willing to colonize all of these grains quite rapidly (<10 days).

I'd love to do a chemical analysis on the resulting mycelia and fruiting bodies at some point to see if they vary in alkaloid content. Anyways, I thought this might help to illustrate how versatile these fascinating fungi are.





Additional Info on Grain and Jar Prep
I do all my grain work largely by feel. That is to say, I don't follow a strict method for hydrating grains. I try to get them to a level of softness such that they are firm, but breakable when applying firm pressure with a finger nail. Here's how I hydrated this particular set (see notes about lids below grain info):

Hard Red Wheat Berry
12 hour soak, drain water, rinse thoroughly with warm water in colander running hands through grains while rinsing, lay out on a towel to dry for 1 hour, fill jar ~2/3 of the way with grains, screw on lid, cover with tinfoil, PC for 90 minutes at 15 psi

Barley
12 hour soak, drain water, rinse thoroughly with warm water in colander running hands through grains while rinsing, lay out on a towel to dry for 1 hour, fill jar ~2/3 of the way with grains, screw on lid, cover with tinfoil, PC for 90 minutes at 15 psi

Popcorn
12 hour soak, drain water, simmer in fresh water over stovetop mixing frequently until breakable by fingernail pressure (check every 20 mins), drain water and rinse thoroughly with warm water in colander running hands through grains while rinsing, lay out on a towel to dry fro 1 hour, fill jar ~2/3 of the way with grains, screw on lid, cover with tinfoil, PC for 90 minutes at 15 psi

Hulled Millet
12 hour soak, drain water, simmer in fresh water over stovetop mixing frequently until breakable by fingernail pressure (check every 20 mins), drain water and rinse thoroughly with warm water in colander running hands through grains while rinsing, lay out on a towel to dry fro 1 hour, fill jar ~2/3 of the way with grains, screw on lid, cover with tinfoil, PC for 90 minutes at 15 psi

Mixed Grains
With remaining hydrated grains from above, fill jar ~2/3 of the way with grains, screw on lid, cover with tinfoil, and PC for 90 minutes at 15 psi

Once the 90 minutes has elapsed, let the PC depressurize before opening. Then let the jars cool at room temperature until you are able to comfortably hold them without burning yourself. Make sure the lids are screwed on tightly before removing the tinfoil and inoculating.

Lids
Two holes were drilled into each lid. The holes were each covered with a piece of micropore tape before PCing. After PC, a spore syringe was used to puncture the micropore tape over one of the holes and inject 2 mL of fresh spore solution into each jar. After injection, a piece of bandaid was used to cover the punctured hole. So, in the end, you have one hole covered with micropore tape, and one hole covered with a piece of a bandaid.

The jars were shaken at about 1/3 colonization and then left to colonize to 100% with no consolidation period (i.e., spawned to bulk almost immediately after reaching 100% colonization).

looks like the smaller the surface area of the grain is, the faster colonization can occur. Not sure how to look at this data beyond that observation.

also saw study that determined sorghum was above and beyond others used in the test, Kizzle linked it some time back* iirc, will edit to add link if i find it.

very cool, is that barley second from the left? looks furthest along in second pic.

had to be a great deal$$ to use media pour bottles, thems not cheap. should hold up to a very hard break up shake and bang on tire for spawning, otherwise tight neck:shrug: good for controlling amount poured when g2g tho!

you drill a hole in lid for GE? what filter? is that MP tape showing on the furthest left?
i wonder if these lids will hold up well with two holes drilled, for GE and LC straw:awesomenod:

damn, great grain masters (g2g) idea, small and super sturdy jar with a forever lid. do small grain, like millet, RGS, Rye in those and g2g to quarts of WBS, wheat, oats (or more of same). lots of inoc points with smaller grains, maybe gives a good leap off for colonizing tub..

got two for agar, the slim pyrex ones, would like to get a dozen these for grain masters and LC if that lid looks good to go after drilled~:dancer:

Yes garlic cut up into pieces and mixed with some other things but it was fruited with success.

*straight RGS (untreated rye grass mix), boiled, strained then PC @21PSI for 100min, above is PM a2g, also have B+, AA+, GT, CB and PE noc'd w/MSS growing out, half of which are a good bit slower. hard to find untreated and pretty expensive tho:shrug:

Yeah, barley is second from the left. It's hard to tell what's colonizing fastest as the shape of the grains does alter the appearance of the myc, and the grains with more surface area in theory have more myc overall. Still, barley definitely performed well, but so did all the grains!

I drilled a hole for GE, covered with micropore tape. I edited the op with more info on the lids. I love these bottles. Really strong and easy to work with. I also like the volume markings because they allow me to standardize the amount of grain in each jar and are useful for LC and prepping agar as well.

Yup! The other grains soften up much more readily, at least in my experience. I'm sure you could do a quick simmer instead of the 12 hour soak to speed up the process, but it's much more hands off with the longer soak and harder to over-hydrate.

(also came up in eat's Powwow thread. convo about millet 24hr soak only versus prepless/ratio method and you confirming here, some only soak, dry and load for PC.

i have completely missed that long soak only prep method up till now, nice mental block breakthru tho!:lol:

Glad to hear it! I think a lot of people overthink grain prep. Once you get a good feel for it, you can easily prep most grains.  :cheers:

This is wonderful, thanks for sharing. I think itll encourage people to take a leap of faith with differemt cereal grains that are cheap to their location.
