Brown/Orange speckles on agar plates

Hi Everyone,

I recently made the transition to using agar, where previously I injected MS spores into grain jars. I prepared some PDYA using Roadkill's recipe and PC'd for 30 min @ 15 psi. I poured 10 plates, inoculated with a flame-sterilised loop and golden teacher MS solution in a SAB. The plates seemed to progress well, and appeared to me to be free of contamination. After 2 weeks, I transferred (inside a SAB w/ sterile scalpel etc.) to fresh plates prepared in the same way. However, a week after the transfers my new plates ALL appear to have brown speckles on them, to varying degrees. The speckles are located solely on the bottom surface of the agar discs, within 1mm of the surface. I am hoping that this is some sort of sediment from the agar mix, as it was certainly not clear when I poured it. I have opened one of the more speckled plates and it has a typical mushroom odour. I have attached pictures of the bottom of one of the more speckled plates and examples of the mycellium present on the top surface of the more speckled plates. Has anybody ever seen contamination appear as these brown/orange speckles? Or am I being paranoid about harmless sediment?

The mycelium looks alright, 2nd picture is it just on the surface of the agar or all throughout the agar?

You said the agar wasn't clear when pored it's likely just clumped agar bits.

If it's not just the surface could be agar not dissolved properly (clumped up)

Thanks for the response. Appears to all be located on the bottom surface of the agar disc. I took one plate and carefully scraped away the very top layer of the bottom surface with a scalpel, and there were no speckles beneath. Nor are there any speckles on the top surface of the agar disc.

Alright sounds like your plates were contaminated before you poured as the specles are only on the bottom surface

I assume you used a 10 pack of pre sterile plates, that package must have been damaged or a pierce in the plastic packaging

Hopefully they're okay, I'm going to keep an eye on the spots. If they increase in number I assume it's contamination, rather than sediment from the agar mix.

They seem okay to me, purely because the number of speckles has not increased and the existing speckles do not seem to have increased in size. There have also been no unusual changes in the mycellium. :thumbup:

Those dots are always on my agar. It's just the ingredients not being mixed 100%. It is fine.
