Well you want enough water it wont all evaporate while pcing. You dont want too much water causing it to splash up over your plates. Elevate your plates off the bottom of the pc,  I used a layer of jar lid rings on the bottom then the rack that came with the p . A little higher elevated is better so u can put in more water without the water going to high up the first layer of plates loaded into the pc.

Primelines, thanks for your response.

Including using jar lids to raise up the rounds.

So... can you suggest how much water might be a minimum?

ive had great results by stabbing a bit from the cotton swab with your xacto knife and pulling away some bits of cotton, and then getting that to germinate in your agar plate. since i always make no pour ones, its better to use freshly made plates since it has more moisture than older plates, thus, making germination easier. with a single swab you can probably make 3-5 or more plates thats almost guaranteed to achieve some sort of germination, given your spores in the swabs are still viable.

edit: I didnt really read the question it seems, what I wrote was for PE and other hard-to-collect-spores swabs. xD

just leaving it there for others.

Depends what size pc you have. I never measure it out exactly. I should though. I just put the rings on the bottom, then the rack, load my plates and add water till it comes up to a bit under half way of the bottom layer of plates. If I'm not doing a shit load of plates, the plates float in the water so I put a dinner plate on top of the plates so they arent floating around. I put the plate as a weight on first then add my water so they dont float.

Can someone please give me the ratios for this recipe, to make enough for 8 plastic minis? Is that what this recipe is for?

Sorry if that was clear in the tek, I was having a hard time understanding how much this was for...

most PCs have a fill line etched in the inside of the vessel.  at least the prestos do.  I always use that as a guide for water.  hasnt failed me in 10years..  :smirk:

It does not have to be micropore tape. I've used polyfil for most of my pasty plates. Anything that can be bunched up tight enough to create a filter should work but polyfil and tape are so easy to get I can't imagine branching out for any reason.

I was thinking using PC sterilized cotton tips, a few dry and a few wet. Then, taking the dry one to rub the spore print to pick up some spore then use the wet tip to "steal" some spores from the dry one, then inoculate with the wet tip the agar plate. This should work i think?

Why are you thinking you need two different swabs? One dry swab is fine for grabbing the spores and Inoculating the plate.

Micropore tape doesn't always work because, per 3m, it is not meant to filter small particles and cannot be sterilized using a pressure cooker. It's really not a great filter. All of my plates have failed using micropore tape. So the answer is that it probably depends on how clean your environment is whether or not you can get away with the tape.

I plan on using syringe filters in the future.

Syringe filters sound overkill, I see many people successfully using tape on the lids with these, I'd think its gotta be getting in there some other way...

Agreed. People had been using MP tape for years. Recently someone posted the package label. Now, MP hysteria has broken out where everyone is freaked out that it's going to contaminate their grows.

It seems to be working for this application though.
