When I get a difficult to germinate swab I cut the tip off and leave it on a plate. Sometimes with an older swabs I have had alot of difficulty and this trick always does the job no need to add sterile water or anything like that.
I have forceps and some steel shears I use to cut but you can use flamed scissors or try scraping tree he cotton off with flamed scalpel may work also. Good luck with whatever you decide to try I hope your spores germinate for you.:thumbup:

No germ from swab

Soo I’ve mostly used prints with agar in the past. I recently acquired some swabs and I didn’t get any germination from the one I tried. All I did was gently swipe it over the agar surface like an inoculation loop.

Is there something I’m missing here? I’ve looked and it seems like other people do the same as what I attempted but I’m kind of confused. Should I be hydrating it first or something or would it be best to go with something like this? https://www.shroomery.org/forums/showflat.php/Number/25162436

Thank you!! I’ll go ahead and give that a try, but how do I know when I should do that with a swab if I don’t know old they are? Should I just go ahead and do that with all of them?

Well if you know it's pretty fresh it's usually not necessary but what I like to do is start with 3 dishes.
I'll swipe 2 and clip the swab end off in the 3rd. And most swabs I use are double ended so I'll save the other end just in case I have no luck with those 3 dishes.
I will then either clean up and test all 3 separate or select the fastest/cleanest of the 3 and stick with that one. Since I'm using no pours it's almost no waste at all.

I use tweezers on swabs and pull off strands, worked on ape swabs I had.

If the swabs are really old I'd go with a slightly looser agar mix to help re-hydrate the spores.
