aquatic antibiotics
    #2623789 - 05/01/04 12:55 AM (12 years, 11 months ago)

I've been reading a little bit about fighting bacterial contaminatants on agar. I've been working on a contamed print and I am now out of GS plates. The cultures are on their P1 after the MS germination and it appears the bacteria is still fighting strong. I was wondering if anyone might know which marine or aquatic antibiotics are gram positive or gram negative. If they work, I'd much rather buy fish pills than mail order a controlled antibiotic.

I think it would be easier to answer if we knew what the actual antibiotic agent is in each "fish pill". I did not know Gentamycin was controlled, I have purchased it from an animal supply house, in injectible solution. I have also seen it on myco-supply stores as well (but is only gram positive).

Do the bottles (or whatever) of fish antibiotics say what the active ingredients are? I don't know when you plan to add the antibiotic to your agar, but most do not survive the heat of pressure cooking.

I did find this, I don't know if it's any help. It lists some common antibiotics used for fish.

I can pick up almost any anti-biotic through a couple of places dealing in plant propagation... invitro culturing is becoming quite popular

I made some trials with penicillin and streptomycin some time back, and those 2 antibiotics are know to not be heat resistant.
Yet, there was some clear indication that they work nevertheless to some extent.

Bacteria are gram sensitive -- not antibiotics. If we are talking about heat resistance gentamicin is the way to go, but Anno found out a while back (as he stated above) that is is effective w/ heat to some extent. If it doesn't work, Mabey try raising the concentration like 1.5x? If not, 2X????

I don't know how the denatured antibiotics could effect the mycelium, though, if it would, at all. Worst case, be sterile and keep isolating, and remember to KEEP THE PH DOWN. (4.5 if you need)

How are you keeping the PH down ?
I tried it once and had the problem that the agar did not solidify, when the PH was that low.

Thanks guys. I was investigating the options because I do not want to order GS off the net if I could buy something similar (but not the same) locally.

Do you think that the insides of the capsules are clean enough to use after autoclaving the agar? How should I figure out how much antibiotic to add? Lastly, with my live cultures that are still combating bacteria, can I add h2o2 and antibiotic?
If I could, that would seem to negate a few contaminates that may be inside the capsules.
