I have seen people suggest their environment (molds in the air, pets, living in an old house, etc...) could be to blame for their contams.  Your environment doesn't matter at all if doing proper SAB work.  If your air is still and you are getting contams in your agar or jars it is because of an existing contam from the getgo or because of your technique!



With your SAB I suggest no smaller than a 110qt tote and make the arms holes big as hell.  This prevents turbulence within the SAB, as well as prevents bumping the edges of the arm holes.



In regards to clean working environment, I am incredibly far from ideal.
I do my SAB work in the same 250sqft space that my cat resides, all my belongings are, 100 year old dusty/moldy building, kitchen and bathroom literally 5 feet away from my SAB, my SAB is no more than 3 feet from my bed, cat hair everywhere all the time, etc... 


How can I guarantee my work will be clean despite all the cat hair, dust, and whatever else?  Simple.
1.  I turn off any circulating air for an hour prior to working.
2.  I soak my x-acto for a few minutes in 70% ISO prior to working.
3.  I wipe my jars/pasty plates/slants with an ISO soaked paper towel in the SAB before opening them.
4.  I flame the blade red hot between each transfer and flame the upper part of the handle every three or four transfers.
5.  I make smooth movements that are least likely to disrupt the air.
6.  I wipe my hands and forearms with 70% ISO periodically throughout session.


Common sense is king in SAB work!!  You can work a dry SAB or you can mist inside.  You can lay paper towels down on the table, or not if you don't want to.  Doesn't matter, the nasties aren't floating around if you're not disrupting them.



Every successful grower has their own ways of going about SAB work and that's okay.  The one thing they do is they all work smart and they all make sure their air is still.  Environment is irrelevant.  Technique is key.

One of the things that reduced my contam rate with my SAB was just chilling a bit longer and giving it longer to still.  A 30 minute TV show or youtube, or the forums *cough* is a good way to distract yourself while waiting.

Yes.
Mine is small at the moment and so a run isn’t that many jars, but a good 20 min break in between each run, with the room air and the box air left to still again, as well as better overall process, has given me no problems and since coming back I’ve had no contams except those traceable to the syringe.

Interesting, thus are you thinking the main airborne vector in specifically unsettled room air, vs unsettled box air?

When I mist I don't give it more than 3 min to settle but that's nice to know I can cut it down to more like 30 seconds lol

Another simple one is get comfortable. I used to awkwardly bend over and try to look through the sab from the top down. Till I decided to use a perfect height stool. Really reduces movements as well if everyone you can grab is within arms reach of you sitting down.

And pushing the limits is good too because I used to think I needed to use way more iso and disinfectant spray and paper towel than needed.

Do you guys use lysol spray?

I find it goes way  way further. I used to have my doubts about it but I use it alot now but use iso for things like my gloves and inside of SAB. I think it's great for the general working area and tools.

Yeah it's easy to stretch out for sure I just don't care for the film left behind on my sab walls after using it for awhile, easy to wipe I'm just lazy
