It's pretty tough, but i always get 25 plates out of 500ml with some left over, and my cultures really seem to like this recipe.

It will still be pretty stiff at 10 grams even if you wanted to make it 12 .. standard recipe is 10 per 500ml

Standard for who? when i first started agar and was buying premixed MEA powder, the directions on the bag suggested just that, around 24 grams for every half liter. Since then ive just gone with 25+5. I will say the last plates i made, i went with 15+10, and i think it may have helped out with that beautiful rhizo u see. Not too sure though, just speculating.

Premixed agar is telling you to use that much with the malt and agar together not 24 per agar .. 20 grams agar 10 to 15 grams malt per 1000 is a stiff mix.. you can do 13 to 13 to 1000ml for a softer higher nute and anything in between

2% is standard for both agar and nutes and depending on the agar brand can be quite stiff. Telephone brand for instance is bulletproof at 2%.

Oh alright, i didnt realize or think about how much of the premixed powder was malt. i just kinda figured it was a really small amount and wouldnt add more than 10 or so grams to a 100 gram bag.
I like the way my plates are with what i use and its just quicker for me to weigh out 5-10g of malt and drop a packet of telephone and call it a day. Just simple things.
Whats the downside to my agar recipe, besides using more than i need to accomplish my goal? genuinely wondering

Probably dry out way faster.. besides that cant think of nothing . Just use 1000ml double your malt and store any left over in a Mason jar in the fridge for later use .. unless you use media bottles and pc then you can just store that in the fridge and microwave when needed again

I do use a media bottle but i only have one 500ml pyrex slimline. Im ordering some media supplies on friday when i get paid, a couple more bottles so i can presterilize and just re-melt in the microwave. Ill try out a lower agar ratio. I can say that i've had a few cultures in cold storage for a while and also some regular temp ones that im waiting for to fruit in vitro, and i dont see any signs of drying out.

That's good .. then it would only be you could at the minimum expand 2.5 times the amount of plates .. it's not like agar is expensive but definitely dont need that much for growth of any kind
Caps uses a ridiculous low amount I think he's doing like 6 grams to 1000ml but it helps colonize faster from what I have seen

Just MEA, 25g agar+10g LME+500ml water
The recipe I have work out to 2g agar to 50ml which would be 20g agar to 500ml but think I’ve mistook the ratio and have 1:1 agar to LME
Is this ok?

At the most you need 2% agar. So for 1000ml of media you’d use 20g. For 500ml you’d use 10g. For 50ml you’d use 1g. You can play around to see what gives the best growth or back down for softer agar when dealing with stubborn spores. No need really to make it any harder.

I was referring to notes there that I actually don’t follow I was right ok I was following the instructions for the agar on the packet, which I is 1100ml to the 20g bar of agar so
Now this was my grain soak water agar I’m making as it was convenient of course...I’ve still to get me LME
I’m thinking the LME will add viscosity? Should alter the agar content down or up the water of just leave when making MEA
SORRY for the questions thanks the help here

20g agar per 1100ml is 1.8% so you’re golden there. Dry nutes like me or df are typically used at 2% as well. Grain soak can be used at full strength or half or anywhere in between...soak water can vary depending on type and length of soak so you can play around with what works best. I don’t think the amount of me really affects the hardness of the resulting media at the quantities we’re using.

Cheers I’m still wrapping my head around this and the condensation problems. Is there a comprehensive guide you could point me to so I don’t bombard the thread with these questions. The search function is litter with crap...any one point me in the right direction?
Greatly appreciated

Pasty and bod both have write ups in their sigs. It’s all pretty straightforward. Pour plates in a stack and ahead of time to help avoid condensation. Cold rooms are gonna create more condensation in plates regardless.

What I did do and give your opinion here was I put em upside down on paper towel with iso to drain it out before transferring tissue.
Sound idea?
Also I pour and pc not pour in a SAB

Condensation isn’t really gonna hurt anything given your technique is good and will likely be reabsorbed. Germ plates from syringe or dirty spore prints might be cause for concern as the water will allow any bacteria that is introduced to slide all over the plate. You may be pouring too hot and your room might be a bit cool causing excessive condensation...so you might try pouring a little cooler.
Your pics appear to be on the right track. Grab xfers early from areas of strong uniform growth.
