This is kind of crazy.

Inventor: Paul Edward Stamets US20180021326A1

Using blue light stimulation in the 280-400 nm wavelength range, precursor molecules to psilocybin and psilocin production can be elicited from the mycelium, primordia or fruitbodies of psilocybian or their close non-psilocybian relatives, to induce the formation of neurogenic compounds, including those that are not illegal, and therefore would not violate statutes in the United States or other countries. The goal here is to employ legal neurogenic precursor, congener or analog compounds related to psilocybin and psilocin.


Induction of bioactive compounds from mycelium through light stimulation is a method that can be used for discovering synergistic neurogenic compounds. Blue light activates shikimic acid production in mycelium, and antioxidant polyphenols, while also stimulating tyrosinase leading to melanization while inhibiting laccase and other enzymatic pathways, simultaneously inducing the production of psilocin, psilocybin and other tryptamines from mycelium in psilocybin active mushrooms and their relatives. Modifying the wavelength (i.e. blue light for 12 hours, red light for 12 hours, per day) and frequency of light exposure, including “pulsing” of the light, can help articulate the expression of active neurogenic ingredients. The shikimic acid pathway gives rise to aromatic amino acids (phenylalanine, tyrosine and tryptophan), which are precursors for the formation of ergot alkaloids, lysergic acid, psilocybin and psilocin (Isaac, Fungal-Plant Interactions, Springer Netherlands, 1992). The inventor postulates that light stimulation evoking the transformation of ergosterol to vitamin D will also influence the pathways where light induces neurogenic compounds from mycelium. These pathways are interrelated and can be used for creating medically significant novel compositions whose synergy is useful for repairing the neuropathic damage by promoting neurogenesis and synaptic integrations. Exposure of mycelium to select wavelengths of light, especially blue and red spectra, may induce indole alkaloids related to psilocin analogues that have distinct antiviral and neuroprotective properties.

No, hundreds of people already have. They are plenty strong as normally grown when grown in the dark aside from shit yields. Show me something he's researched or done with efficacy. Host defense is made from mycelium extract and propped up by his own "research" that mycelium is full of the goods. Despite all the research saying fruits are far more medicinally useful and that also most mushroom medicinals are pretty overblown anyway. I personally think it's distasteful to say mushrooms cure cancer and give people false hope. His mother's doctors said she was cured from her treatment and had no idea Paul fed her mushroom extracts. Maybe it's because he's a tinfoil hat man and there's a cancer conspiracy... it's intellectually shameful to go about doing things that way then reporting on them as miracles. Laypeople eat it up.

Bod, I'm with ya on the Cancer stuff It's fucked up. Help me with this, he has stated a few times that UV is needed to trigger biosynthesis but I can't find anything to back that up. The closet I can find is in the use of HPLC-UV for detection.

Every pathway ive seen doesn't show light at all as a step or even a catalyst for any of the steps in biosynthesis

From personal experience having a family member (my dad) that started using Pauls products (not sure which one) after being on radiation for a few weeks and was told about it from a fellow patient (not as a cure) it doesnt seem to be snake oil really imho. His antibodies and other immunity factors had improved dramatically along with his symptom tolerance just a couple dr visits after starting it for its natural anti biotic properties. Even his dr said that polysaccharides in mushrooms can help white blood cells and overall immunity And i don't think Paul ever said it cured her cancer but that it saved her life by boosting her immune system that greatly needed improving as she was undergoing conventional cancer that people very rarely recover from such a chronic stage. Plus the research hes doing with the bees is super solid and beneficial and from what i here is promising and going to be released to the public to help apieries for free.... I love Paul lol sorry

I also don't understand what hes getting at with the vitamin d-psilocin analogues but i think he means that the phototropism reactions mushrooms use blue light 320-400nm to induce vitD and fruiting are tied to psilocybin production. I remember him referring to an old experiment showed that p cubensis mycelium doesnt produce psilocin unless exposed to far blue/uv light. But idk what that is clipped from.

I'm not a Paul hater, I have learned so much from his early work. But Bod might be like me and lost some one close to Cancer. It's a touchy subject. I'm interested in what you dad was taking and how much. Can you please find out and let me know?

I see a lot of big flowery words, many claims and presumptions but very little evidence or research.

For as long as the whole “blue light increases potency” thing has been around you would think someone would come up with a solid study to show it.  I believe in recent years there has been one study that showed many mushrooms enjoy blue light, but nothing to suggest it has anything to do with psilocybin production.

On the other hand we do know it has little to do with psilocin production and if it did that would be bad, as the pathways discovered show phosphorylation first and if we were going in steps, it would be best to call Baeocystin methylated norbaeocystin and psilocybin methylated baeocystin. So if blue light is causing psilocin production, it’s causing the dephosophorylation of psilocybin, which is bad and if the phosphorylating enzymes find it, they will spend energy to turn it back.  Increasing psilocin production seems like a potential stress to the mushroom and not a benefit.

Additional that if it’s being produced in a manner similar to vitamin D or melanin we should find a higher concentration at the surface, just like we do melanin, and a more average concentration in the core, as not being exposed to light the core doesn’t need to tan.

I know lions mane was one, particularly whole powder in smoothies. And i believe one of the earlier blends paul had. I will ask he has started growing his own reishi, lions mane, oysters and is having me help setup a wood pile with portabella mushrooms. He also stopped eating meat or preservatives. I still do all that horrible shit hes like the world's strongest man compared to me lol and hes 72! Im not contributing it all to the products Paul has but they do help and i think its significance is great enough to cause a lot of people to look into what other ways they can clean up their intake and start seeking better living habbits as they realize the benefit in them. It was like the first good news he heard and i feel it helped him just in that regard alone to keep pushing. But i do understand there is nothing worse than hearing someone promote any product as a cure in any way even if it does dramatically help a subset group of people. Because the expectations of most will be crushed. Similar to how they promote CBD as a cure all yet even for what it's best at seizures it only decreases epileptic attack frequency and or intensity in less than 40% of epilepsy patients... But i also see how its hard to market it without giving false hopes yet still having enough incentive to buy it or grow it eventually so it gets in as many peoples hands as possible..

Originally i was like "no way" then i read it now i got 50$ of B-Roids in the mail thank you ebay. Unfortunately they call for an insane amount of 5g per liter of substrate so that is expensive but not too expensive to test it!

I’ve met Paul. He’s a down to earth guy that is literally obsessed to the teeth with all things remotely mycology related up to and past his ears, radiating into fucking space...and he’s not stopping. He’s got a lot of science to back up what’s in his books and that’s going back to the 70’s. He emerged out of the company of people like Weil, McKenna, Pollock, & a bunch more of the classic early psilo-folk type people like that. His latest “Fantastic Fungi”, is just beautiful, and apparently has a film complement that has been in the works for over a decade. The trailer looks great anyway. I also know he’s been contracted for work by the US government for something called the Bioshield program (some anti-terrorism related endeavor) specifically for his knowledge on fungi as pertains to warfare/defense.

I’ve never bought anything from him except books indirectly over the years, so I cannot speak personally regarding his products, nor would I if I had, since I do not posses any remote way to even begin assess it, whether it be people/peers, technology, or what have you.

Yea started over after years of just doing a cased grain jar in my dresser once or twice a year for headaches. Recently i decided they need to be my new old hobby and i plan on getting a new flow hood soon but going to start working agar in a SAB once i get my first flushes of PE, PEU, APE, AA+,Huautla, and p. tropicalis. Also plan on starting subaeruginosa to have chips in time for next spring.


But yea I'm interested in seeing small amounts of the brassinosteroid are effective enough to warrant use it is possible the amount they use is unnecessarily excessive since they only tested one concentration of it.

Hey you guys only need to add gas that costs $20 a gallon to get to 37 mpg. Hey well i get 32 mpg on $4 a gallon gas. That's like this additive. If it doesn't even pay for itself in returns its pretty dumb imo

I totally agree looking at the prices available with a quick google search its insane but i think the lower amounts of it will probably produce statistically significant difference in speed and pin set. It would have been nice if they didn't do invitro fruiting or if the did something like a grow bag etc that way we could have seen the fruits unsquished. Also curious if would help slow Panaelolus species fruit within a month from spore! Then it may be worth the cheddar lol when i get it i will start a new thread and post the link here. Tho im at least 6 weeks from having an isolate that would be suited for side by side. Started fresh just 3 weeks ago.

So I should use a LED bulb with 6500K.

I have a 5000K LED, but the 6500K lights have more blue, so will probably produce more psilocin.

Abstract
Different variables including species, strain, glucose and ammonium succinate concentration in
the growth medium, pH, temperature, timing and oxidation have been accounted for the Psilocin
(PC) and Psilocybin (PB) content in the "Magic Mushrooms"(MMs). These are but some of the
variables in a constellation of factors that complicate consistency in the production of PC and PB.
In an attempt to study the effect of light on chemical constitutions, some samples were kept in
dark, some samples were kept in dim lighting, whereas others were exposed to natural but indirect
light. After picking and drying Mushrooms, a simple one-step extraction involving
homogenization of the dried fruit bodies of fungi in chloroform and derivatization with MSTFA
was performed. The samples were then analyzed by gas chromatography–mass spectrometry. This
investigation shows that the relative PC and PB content of the mushrooms is highly dependent on
the light condition. This variation could amount to 100 fold of active components (PB and PC) in
samples harvested in dark condition compared to the samples harvested in indirect light condition.
Therefore it can be concluded that UV light may have destructive effect on the active components,
which would readily explain why sun-struck collections are less potent

Using blue light is supposed to help a lot of species in certain stages of growth anyway right, i thought this has been tested repeatedly (some species do prefer other wavelengths like reds apparently).
And what color was it that stimulates production of cordycepin in Cordyceps compared to other light?

If light factors in it's not a "catalyst", but there may still be a correlation about certain metabolic processes or growth development and their induction by light as a trigger... The biosynthesis of relevant alkaloids may very well be indirectly dependent on those metabolic processes, but it wouldn't necessarily show up as a known critical factor in literature.

Obviously we like to see more evidence etc but it's not at all implausible. This was in a patent right, so how is it substantiated?

AFAIK his mother was believed to bounce back much quicker from radiation wiping out her immune system, from turkey tail stimulating the immune system. Something which apparently many mushrooms may do because or compounds rather common in them (tho those are not created equal). Not the same as curing cancer, but I'm sure he blew it out of proportion talking about it here and there.

People say 'at least he knows how to sell it all'.. :\  trust me I am skeptical about the dude too and it feels more like shilling or something. I'm also fine believing that he likes to take credit for other ppl's work in some way.

But not sure it's such a good idea to be biased about every concept that came from his direction. If its bad, just take it apart for reasons. If he doesn't offer nuance or fairness, maybe that's what we ought to try and do.

Anyway I think he is playing a tricky game. Someone might argue that it helps get fungi good publicity, but if he oversells certain badly founded claims and there is backlash it may harm the reputation of the field of research.

But when it comes to scientific claims let's stick to scientific validation or dismissal.

I run blue light and get good results. It’s not always 12 hour cycling...this amount isn’t 100% needed, but it’s not going to hurt anything to mimic natural patterns of sunlight like that either. Substrate sort of just pins when it pins; a drop in temperature, increase in evaporation, some type of light source all seem to help do the trick if full colonization is reached. I’m sort of stating the obvious here, but still.

Some supposed studies suggest that milliseconds is all it takes. I honestly am not sure how we get this 12/12 cycle.

Blue light will work even if it's just a part of the cool daylight spectrum which otherwise may be used. When i got into blue it was because it lacks the unnecessary wavelengths so is just a fraction more sustainable, i didnt know how many other's caught on to the idea. Or how fucked up you get from looking into it.
So honestly I consider it a novelty now, unless systems are implemented to change the lighting depending on the presence of an employee/worker.

Just like some other things like the use of an incubator, at a very small scale it gets kind of like a novelty or concept.. or just kind of silly or experimental. While if upscaled there can very well be a point and it may be more worth it.

There are no actual mushrooms in Paul's supplements, they're based on grain/rice/mycelium biomass and contain mostly starch.

Just in case you don't believe what I say about it. Take it from Paul Stamets:

Here is an excerpt from one of Stamets’ patents:

EXAMPLE 9 [00123] The medicinal mushroom mycelium is grown utilizing liquid culture techniques. Whereas growing on rice might have 30-40% conversion of rice to mycelium, liquid vat culture may have essentially complete conversion with >3x more mycelium per unit mass.

30-40% conversion of rice to mycelium. Meaning 60-70% rice.

Check this research that confirms that 100%.

High alpha-glucan = starch which is from grain. While they will tell you the mycelium consumes the majority of the grain, the actual test data tells an entirely different story.

That is the reason they do not share actual lab test reports of their products. The same with Paul Stamets's products. Lot's of talk, but zero facts.

Their products are also not extracted, meaning the bioavailability is unpredictable for 80% of people.

Summarising: you are paying top dollars for useless starch.

IF you want to buy REAL mushroom supplements for your dad, I've only found 2 companies that actually SHARES! lab results. Very low starch content and high beta-glucans. They tell you the % of Beta-glucans. This is the second company! they use only fruiting bodies and they tell you exactly how much starch content there is in their products. Although, fruiting bodies are not where the most medicinal content is but is certainly better than Paul's products with 80% starch content or more.

for some mushrooms the most important active ingredients are actually in the mycelium (Lion’s Mane (erinacines), Turkey Tail (PSP/PSK), Cordyceps CS-4 (no fruiting body extracts exist!))
there is a difference between mycelium-on-grains/rice (biomass) and pure mycelium. Biomass should always be avoided as it is contaminated with undigested substrate in the form of starch. This includes all US-cultivated products. We only use pure mycelium, free from starch, if the products properties request the use of mycelium
quality claims are only worth something if they are backed with actual test reports. This includes a claim like 'fruiting body extracts are better than mycelium-based extracts' or vice versa. Nice statement, but where is the proof ? Also see our 'I'm confused by all the information online' entry, above. Using ‘we use fruiting bodies only!!’ has the subtext ‘it’s better!’ but actually only reveals the ignorance of the vendor, and the actual specifications, if any (% of glucans, terpenes etc.) usually have no verifiable source or backup in the form of official documents. Only specifying active ingredients is acceptable as proof of superior quality.



Going back to fruiting bodies vs pure Mycelium for medicinal content.

Research has found that pure mycelium has most of the medicinal content! Companies like Oriveda uses a method called deep layer cultivate mycelia which is basically what we call "liquid culture" but they grow pure mycelium in MASSIVE amounts and extracts the medicinal content from there. NOT GRAINS. Companies like Paul and Aloha medicinals just want to sell you starch, is a FACT. They only care about money, not curing your mom or dad, if they would care they would share LAB TEST RESULTS.

So what exactly are the differences between the fruit bodies and the mycelium? Would psilocin and psilocybin grow in mycelium as well as the fruit bodies, or is it only produced at maturity?

They are also produced in the mycelium but a little less of it. Depending on the species though, it can apparently be viable to extract the mycelium for the alkaloids, possibly worthwhile.
I kinda want to assume the mycelium reacts the same way to light where growing in darkness gives more psilocin and growing in light more psilocybin relatively (they would be converted into each other, im not talking about a change in overall potency).

But my question would be: how local is this effect? Are these concentrations (proportions) changed throughout the mycelium - even the bottom dark parts - if the above ground parts exposed to light are receiving photoperiod triggers?

As far as i know psilocybin is produced in the mycelium as well... I don't know in what concentration tho... fruiting bodies might have more psilocybin content than the mycelium i don't know but in Medicinal mushrooms like Tukey Tail and Lion Mane the mycelium have much more healing properties. Just don't grow it in grains :P it will result in high starch content.

Fine and dandy if you want to grow with one spectrum. Most people just use a fluorescent which has more than enough of any spectrum mushrooms need anyway. Besides that article really focused on correlations to plants which makes me think it was just some people who were grasping for straws to publish another paper to keep money rolling

I’m not a fan of pauls, however aloha medicinals also claims that mycelium is full of goodies.

I’ve yet to email aloha for their analysis but I do believe aloha will provide data. Paul’s company will not last I checked, as they believe it is basically inconsistent for the lack of remembering how they put it exactly when I emailed them long ago

I personally am a fan of fruits and myceliated grain supplements. I’ve eaten supplements and definitely noticed they were cleaning me out. My nose told me. Not to mention a lady who swears they cured her digestive issues to say the least which I’ve met first hand. In fact I recommend she try them.

To be clear the supplements I was eating when I noticed a difference was ohm brand Paul’s are pricey for sure. Make your own try them out for a month see how feel then come back and report. My conclusions were made long ago. Seems to be a lot of back and forth on this topic and many haven’t even spent $20 on some supplements and tried them lol

Someone did a study on the components of common mushroom supplements. Paul's did the worst. It's mostly starch. And the polysaccharides in the product not extracted so they're barely bioavailable

Both companies won't share test lab results. Both won't share the starch content. Both companies lie in that aspect. And many other companies not only those two. The only two companies i found that share lab results are the ones i mentioned above: Oriveda and realmushrooms

Is really shocking knowing that only two companies share actual lab test results. I admire Paul and John Holliday, they know their shit, but their products are mostly starch and that is a fact according to the study i mentioned above that verifies that 100%.

Read what Oriveda says about their turkey tail extraction and cultivation method. They use a technique called Deep layer extract, basically they grow the mycelium in liquid cultures, in massive sterile tanks of liquid culture.

According to the original research the most potent source of 'PSP' is that COV-1® mycelium strain (more details here)  which should ideally have been cultivated using so-called 'deep layer cultivation' (growing it in sterile laboratory tanks, in liquid substrate). This technique is standard in China but hardly used anywhere else, due to the high cost.

Cheap cultivation techniques are using rice/grains to grow mushrooms (solid substrate), but since it is impossible to isolate the spiderweb-like mycelium from the substrate in the end the final product is in fact 'biomass', not mushroom. It is contaminated with undigested substrate in the form of starch; levels can be as high as 80% (!).

Such products are not per definition without potential but in general their actual therapeutic effects are low and the value for money is poor. On top of that these products are in general not extracted, rendering them even less useful. They do not specify any bioactive compounds on their labels and a 2016 test report showed several of these product contained no beta-glucan at all, only starch. Research found that the immunological effects of mushroom extracts are up to 10 times higher when compared against non-extracted powders.

This is what they say about their Lion mane, they do use the fruiting body of the lion mane and also pure mycelium. They use both for the extraction, since there is a medicinal compound that is only found in the fruiting body.

If you buy a Lion's Mane supplement because it lists a high level of beta-glucans (or polysaccharides) you are not spending your money wisely. It makes more sense to choose a Lion's Mane supplement that specifies/guarantees Lion's Mane-specific compounds, right?

As said, in Lion's Mane the quality markers are 2 specific groups of bioactive terpenes; these contain the NGF- / BDNF-inducing compounds in Lion's Mane. Those compounds are known as hericenones (present in the fruiting body only - mero-terpenes) and erinacines (only found in the mycelium - di-terpenes).

Until now over 30 of these compounds have been identified in Lion's Mane. It is impossible to test for all of those individually. With only a few exceptions there's no standardised reference material available.

Quite a few research papers however describe in great detail how these compounds were discovered, isolated and quantified. The independent third party lab we use used this information to come up with a reliable way to measure / quantify these compounds. Check our COA. This level of detail makes L+ unique.

What also sets Oriveda's L+ apart as a unique product is that it is the first and only Lion's Mane alcohol extract. Only an alcohol extract contains a noteworthy amount of these compounds - they are strictly alcohol-soluble. When we were developing L+ we have tested dual extraction as well but this would have meant an unacceptable compromise. Levels of alcohol-soluble bioactives were low. In particular the mycelium had a very low yield of less than 0.1%.

Then there is the fact that Oriveda's L+ contains both fruiting body and pure mycelium. There are no other Lion's Mane products on the market containing pure mycelium, which is a major -and surprising- omission because the mycelium contains the most important NGF-inducers; erinacines (their NGF-promoting effect is up to 4 times stronger when compared against hericenones according to research).

The mycelium used was cultivated in liquid substrate, formulated using the latest research in order to produce a very high level of erinacines. It is the first time this technique was used succesfully outside a research facility. The results were excellent - at least 2 % erinacines are guaranteed.

After repeated alcohol extraction (to squeeze out all active compounds) the final product is 100% uncompromised Lion's Mane extract, free of additives or flow agents.

It should be obvious Oriveda's L+ is a better choice than a liquid alcohol 'tincture' because in such products the main element is alcohol, not Lion's Mane. If you would allow the alcohol to evaporate the remaining deposit equals maybe 1 gram of powdered extract. Such products also do not / cannot guarantee any bioactives. In the case of the Turkey tail and i guess Reishi, the medicinals compounds are found mostly in the mycelium not in the fruiting bodies. I mean you can also found some of the medicinal compounds in the fruiting bodies just not as much as in the mycelium.

That's terrible.

Wanting to catch a profit to be sustainable is no shame, but if these guys sell products which they should very well know only basically contains actives enough to be allowed to put it on the label and not actually caring about bioavailability and concentrations... then it can harm the whole field where there is still a lot of promise.

Too bad there is not a public forum per se where they can be challenged for these findings.

Idk if this is just the absolute typical thing to happen for a company set up to maximize profits whatever the cost, because then sure if they can take advantage of the fact that few people could really challenge them.. it would encourage deception.

However if it is just about sharing analysis results: maybe it is reasonable that when these companies feel they give too much info away, they react defensively. Not saying it's not suspicious, but maybe it is too much for make assumptions too.

That said: seems it is super super common for companies to screw around with alpha vs beta glucans etc. Maybe they also feel justified as long as it is a complicated field of discussion, but that would obviously be making it easier on yourself to try to douse yourself in the same ignorance.

thats a shame...

maybe a little expected in a field where a lot of people seem to act primarily on beliefs rather than facts. They want to just believe it works and its just good stuff, and rather not get deep into the tricky reasons why there seem to be a lot of prerequisites for reliable activity.

Do you guys have some links to studies establishing this lack of bioavailability related to either mycelium and/or fruit bodies?  I'm questioning everything these days.  I tend to agree with Bod regarding the "suspect" nature of Paul Stamets, I think he means well, but he does give off a charlatan vibe to me, I question the rigor of his science. 

That said, I've seen some youtube vids from people who claim to be experts saying you have to do extracts to get the medicinal value, but people have been eating mushroom fruit bodies for medicinal value for thousands of years (especially in Asian countries).  I'm pretty certain an extract is not required for medicinal benefit, but I could certainly see a case whereby the medicinal value increases after an extract (but how much? and what is lost in the extract process?).  I want to see reputable, controlled, peer reviewed science on this subject. 

Then of course there is also the problem of making sweeping generalizations - just because one specific compound is more bioavailable after an extract doesn't mean the same applies to every compound found in various mushrooms.

Regarding light spectrums, I have found that just using a full spectrum light works great, and LED clearly beats fluorescent bulbs in this department.  Even cheap LED bulbs work great.  You can actually easily test and see this for yourself, here's how:
