Question about cooking agar in media jars
    #26167475 -

So in the past I have been using a glass bottle with a long neck and a synthetic filter in the lid with a hole drilled out. This worked good no contams so far. I just got my 500ml media bottle with lid and I'm wondering now if I should drill a hole in lid and put a filter in it or if I should just loosen the cap when I pc it. From the posts I have read people have been just loosening the lid then waiting for the pressure to drop to 0 before tightening the lid of jar. My issue is i dont have a pressure gauge on my pressure cooker so i don't want to chance my jars sucking in bad air when it forms the vacume. Does anyone else have a pc with no pressure gauge and prep there agar in media bottles ? What would be my safest bet filter in lid or loosen lid?

If you don't want a vacume to form, wait until the pc cools.  Not just down to pressure.  It's the change in temperature from hot to room temp that makes the vacuum.  Just wait until it cools.

Not sure about the filter though.  I'd use one just to make sure.

Well I thought the vacume happened once it cooled down it goes from positive pressure to 0 then down to negative pressure then when you open ur pc it sucks air in and if my lids loose thats what I'm worried about

Only a sterilizer pulls a vacuum. A pressure cooker usually will not form a vacuum. The overpressure plug, safety popup, and weight all tend to leak air back in after it hits zero psi and starts to cool down more. As soon as the safety popup drops you can open and tighten. You can usually hear that even stoned a room away

What Bod said.  If you follow his EAF agar tek you’ll unscrew the lid a little before PCing so there’s no pressure differential during the PC cycle.  Then, tighten and shake after you get it out of the PC.

I thought they were talking about the actual container itself.  If tightened too early before cooling, a vacuum will form if not vented.

Not the pc itself.

Vacuum will happen if you PC with the media jars lid screwed on tight.

...I think..

All I've ever done is PC, open at zero psi, tighten and put on the shelf to cool:shrug:

Only if the internal pressure escapes during heating and outside pressure causes the lid to seal the vacuum on cooling. More likely IMO the bottle will burst.

Media bottles are usually not rated to be pressure or vacuum resistant. They are available, but not as a standard feature. Even those shouldn't be autoclaved with the lid on tight for the same reasons. They are generally rated for 1.5bar (~22psi). If the PC undergoes a rapid loss of pressure(blowout plug, seal failure, etc), then it could very likely cause the bottle to overpressure and burst.

I do. With media bottles I loosen the lid about 1/2 turn and cover it with foil. Then when the cycle is done I put the PC (little 6qt) in a cold water bath to get the pressure down faster. No problems with boilover yet even with leaning 500mL bottles over so they fit under the lid.

Yeah, just leave the lid a little loose.  Should get enough air...

I leave the lid loose with foil, after the pc run I either let it cool completely before moving to the Laminar Flow Hood or cool it in front of the running Hood, I haven't noticed a difference between these two methods and don't believe air is sucked into the bottle during cooling, if it did I would certainly have contaminated dishes especially when I let it cool in my kitchen rather than in front of the Hood.
