It's not scientifically proven yet.

Those cacti might produce mescaline as defense mechanism agains predators. That's why it is concentrated in the outer green flesh, less in the center of the plant.
A cactus feels when it's laying on it's side. After some months roots start to grow all along the side underneath the plant.

Now it can be that a cactus, which is laying on a side for a long time, produces more alkaloids due to more surface area beeing exposed to the grund. (As most predators are small animals, more parts of the plant beeing close to the ground means more attack surface for those predators)

I have read that cacti get stronger over time when laying down several times now.

As a fact, my strongest cactus trip was from as little as 8 inches of a pretty thin cactus that was laying on it's side for almost a full year. !
It has grown roots all along the side, had the most bitter taste of all cacti I have eaten, and blew my mind completly. As strong as 250mics of lsd, almost too much to handle. Very intense visuals.

Sadly I haven't tested said cutting before laying on it's side for so long, so I have no comparisons of it's potency before and after. :shrug:

I've seen a great deal of commentary on "darking" a cut prior to use as being a pretty reliable way to potentiate the alk content-- I have also tried two cuts of the same type acquired at the same time-- the first I made right away, and it was pretty weak (say 4/10 strength) but I let the other one (clearly identical type) sit in the dark for 7 weeks prior to boiling it, and it was pretty fucking strong-- say 7/10.

Grant that they were not necessarily from the same plant-- so there's always a question there-- but I would always dark a cut for at least a few weeks prior to taking it-- because why wouldn't you :wink:
