Antibiotic Agar

Wanted to make some antibiotic agar.

Pretty sure i stock a few things in my bugoutbag. Thnk i gotta Z-pak, some clindamyacin, and some cefalexin (however they are spelled).

Anybody ever use any of that?

I use antibiotic agar all the time.

I just use a pre-mixed powder from a vendor tho. I highly reccomend this way as it takes out a lot of guess work.

It may seem a tad more expensive to go the premixed route but the ease of use and peace of mind are worth it. Plus, if you lose a bunch of cultures trying to fugure out your own mix, how much are really saving?

Speaking of which this reminds me that I need to order more since i'm running low. Thank you.


Pre-mix from a vendor for the win! Anyone want to argue with me on why fucking with some anti-biotic pills is a better idea?

Believe me, I've got nothing but rebuttles....

That's a new word for me!

The term "bug-out bag" is related to, and possibly derived from, the "bail-out bag" emergency kit many military aviators carry. In the United States, the term refers to the Korean War practice of the U.S. Army designating alternative defensive positions, in the event that the units had to retreat.

American english, evolving from the American experience of war.  Non stop war.  War after war after war.  Afghanistan is going on nearly 20 years now.  I don't know how you guys can take it.

Anyways...I use this in my agar.  I saw a post about it here on the shroomery.  It was easy to get, unlike gentamicin.

I'm just wondering what in the world y'all need it for. Ive cloned wild shrooms and grocery/market ones and I've never used antibiotics once for mycology purposes ever

But it's not though. And using it for piece of mind is quite literally the worst possible way to use antibiotics. Using antibiotics when not necessary is misuse because of well known consequences. Like breeding resistance

All the ones that we use that survive sterilization ain't that good. They can suppress bacteria to the point of having a hard time seeing it without killing it.

A minuscule amount of antibiotics for spore germination plates, or maybe all your agar plates is pretty much how it's used.

Doesn't seem like much.

Compare that to the backdrop of antibiotic use in commercial livestock operations, which is where Ive heard most of the concerns of overuse referenced.  That and medical practices of the 20th century where it was handed out willy nilly or something.

The shit I use is approved for pet fish in aquariums.  Sold in a completely uncontrolled way.  The powers that be aren't concerned, not that that means much of anything, but all I have to indicate a negative in all this is your post.

Are you sure you're not just a purist or something?  You really think we're breeding superbugs in our closets and basements?

Second point was that they don't work anyways. 

I'll  leave it to the user to determine that.

Not breeding resistant bugs in the sense of creating some epidemic but more just problems for yourself. They have middle school kids demonstrate antibiotic resistance on petri dished using pads soaked in antibiotics placed on dishes inoculated with non pathological bacteria and eventually obtain a culture that's developed resistance and will grow around the antibiotic pucks.

Ahh... I had forgotten that it wud go through sterilization and that might be a problem for most antibiotics.

So it's not jst as simple as add some clamydia killer to your agar and BAM! you got little to no bacterial contamination, eh?

Holy shit.  Thanks for linking that video man.  One of the coolest videos I've seen before.  I've never seen antibiotic resistance represented like that.  Crazy to watch some principles of evolution take place at lightning speed

So it does kill bacteria? And what it doesnt kill, it supresses?
If that be true it kinda sounds like a win/win.
Is this true? Pls clarify. Im reading shit differently now that im loaded and calm.

In the video the bacteria have a section of agar to grow without antibiotic, followed by areas with low concentration antibiotic building up to high concentration. The antibiotic-free zone allows them to grow to trillions of cells, where some of the cells can develop some resistance and start growing into the low concentration zone and flourish, and eventually cells with high resistance are developed and move into the high concentration zone.

I'm a bit out of it at the moment, so I'm not great with words, but if they inoculated a high-concentration AB agar plate (without any AB-free or low-AB zones), the bacteria would likely NOT grow at all. If the cells cannot divide and replicate in a nice little safe zone first, they cannot develop resistance via natural selection or whatever.

This is one of the principles behind AB resistance in clinical use (aside from the flagrant over prescribing of AB's by doctors). If you don't take your AB prescription regularly as prescribed and/or fail to complete the full course of treatment, AB resistance may occur because the remaining bacteria are given some degree of respite from the antibiotic where they can resume replication and eventually develop resistance to the low levels of antibiotic remaining.

Moral of the story is, if you use an antibiotic use it at the recommended working concentration for agar (can google this for a list/table), and use it for at least 2 transfers of plates in a row to ensure the bacteria is completely eliminated from the culture. Using too low of a concentration is worse than not using an antibiotic at all.

Many antibiotics (if not most, I don't know) don't outright kill bacteria, they "suppress" them in the sense that they cannot replicate or function. For all intents and purposes, this is as good as killing them in the context of mycology. The mycelium will outgrow the suppressed bacteria, allowing you to take a clean wedge of mycelium at the leading edge. And to be safe, this most-likely clean wedge should be transferred to another antibiotic plate, just to be sure that if any suppressed bacteria caught a ride on the wedge, they won't grow after the transfer. From there, the culture will be clean.
