A few questions:

1) To my understanding, even if I take tissue samples and place them on agar, I don't necassarily have a gentically identical clone, correct? Can you tell me what this is called or link me to some literature on this? How can I confirm or deny that my clone is truly a clone (i.e genetically identical)? I'm asking because I purchased lion's mane today, for the purpose of cloning on agar, and the vendor mentioned purchasing spawn from North Spore, so I'm likely to have a good set of genetics. Only $3 for the sample too :-)

2) I started my oyster and shiitake grows by cloning supermarket samples. I see companies offering selected varities picked for performance and such, which is nothing new, but I'm curious: can I get close to this? There is nothing different to selecting and isolating genetics with gourmets than with cubes, right? I personally don't see a reason to pay a premium when I can just do it myself, but perhaps I can't without a real lab (i.e, not a SAB). As a more extreme example, if I want shiitake that fruits on straw, could I realistically produce that as just a regular guy/shroomerite (not a Mycology PhD guy with a lab)?


3) I will be nocc'ing up some agar plates with lion's mane tomorrow, any tips beyond regular sterile procedure practices? I heard it doesn't look quite right on agar and doesn't take well to it.

Also, I'll be starting a grow log here soon! Should I do in the gourmet section or in the log section? I'm rennovating a small outdoor shed for fruiting and bulk sub colonization.

I can't say a tissue sample will guarantee the exact same genetics, but it should: How To Clone Mushrooms.
Multi-genetics from spores will require you to isolate-transfer, and sometimes quite a few times.

The best method for spores, is to isolate growth on an agar that mimics, or is made from your end media.
The end media in most cases will be grain + wood, wood + bran, this requires good knowledge.

If you don't know much about, pH, nutrients and enzymes, use the clone.

Wild idea: certain molds and bacteria induce defensive reaction in many mushrooms. What's mechanism of this? If it's simple chemistry ("presence of compound X means there is an enemy in substrate"), killed bacteria/molds can be used to force fruiting.

Just thought I’d post this here.
I can’t get the pics of the mushrooms that I cloned these from but it’s a wild Michigan clone, notice the white-violet spores! If you are curious, I have the original wild specimens in my gallery I believe

Very nice, Golden oyster is an excellent learner. They will give you signs, at the moment they scream: More oxygen, humidity, water and light please.
In medium FAE (fresh air exchange, oxygen), oysters stretch towards oxygen sources, in low light they produce little colour.

Here is a medium FAE grow, with limited light, but good humidity and water.

     

Now the same block, but outside, with lots of oxygen, light and rain.

forsure, my grow was done in a closet and a fan giving a small bit of fae and not a whole lot of light either. Still was way fun and really different than blue oyster

Yeah I guess my favorite photo of the first flush wild specimens isn’t in any of my shroomery stuff, oh well. Any ways, I’ve always admired the oysters in your sig, it’s so appealing lol

the mycelium has colonized the wbs fast (5 days). I don't see any obvious mould or green shit. I might just open it up and if it smells ok spawn it:shrug:

Well, just thought I'd show my ass again in here. I tried to grow some oysters basically on bucket tek'd HWFP. I used the freshcap 5-gallon grow method. I inoculated it with Mediterranean Oysters spawn on oats at a 1:5 ratio. Seemed to be going fine for about 3-4 days then some of the HW started turning a darker/wetter looking shade of brown in parts... then spreading and eventually the myc production slowed down and stopped... pretty sure it went bacterial :sad:

Anyway to identify if it was the wood or the spawn? I used this type of HWFP (it was sold as such, even though they HWFBricks). They're about 3.4# per brick... because I'm stupid and don't know the difference between wet weight and dry I hydrated two bricks... and I ended up only using 1/2 a brick :uhoh:



I did this kinda seat-of-my-pants, so it's possible that something just went horribly wrong because I was just "fucking around," but I thought I'd get any input possible? Thanks, trying to identify and fix my problems.

The issue with reusable containers is cleaning them in between uses. I tried it out in the past and had higher contamination also the substrate shrinks over time and you start getting pins inside the bucket. I recommend using a bag or something with straw.

I think you have cobweb, it can be very rapid, and wispy. Lots of weird colours going on also. Not 100% sure.

Hey guys,

I've had to move in with a friend and after what I have in blocks are done I won't be growing gourmet for a while.  I would love to share wedges of what I have though:

Oysters:  Yellow, blue, pearl, king
reishi, shiitake, LM, enoki

If anyone is interested PM me.

Also, what is the best way to keep these long term?  They are great cultures and don't want to lose them, just not sure when I can get to them again.

Post a pic, sounds like it might have been over hydrated. That freshcap method looks kinda wack I would make bags of sawdust spawn using lipa tek(basically bucket tek butnin a bag and with pellets instead of cour) then use that to inoculate buckets made with:
lime treated (cold pasteurization),
properly pasteurized chips
fermented chips(cover with water for 2 plus weeks untill they start bubbling, drain for a couple hours)


Tbh bags are the easiest to deal with, then once you get a couple flushes you can use them to inoculate buckets or outdoor patches.

Even if your oyster bag is highly bacterial it will still fruit, just open the holes up and let er rip, I’ve seen bags half green in my discard pile still kicking out fruits. Oysters dgaf

Look into slants for long term storage Sunny. People break off little sticks into them to add longevity to the whole thing.

I'll take some pictures to post soon. I'm only using the bucket part to hydrate the HWFP, sorry for the confusion! I then squeezed them out (field capacity... :lol: for wood?) and loaded them into used/washed myco-bags at a rate of 1:5 with oyster spawn on oats.

Whatever the issue is, it stopped colonizing a while ago, so I've kinda given up on it. I've heard oysters dgaf... but seems like my shit is bad enough to give a fuck :rofldrunk: I've read that some HWFP may be treated with things like oil to be better to burn in stoves... wondering if this stuff was treated with something.

It seemed like the same shit as lipa tek, but since these are HWFBricks I thought that the bucket would be more usable since I couldn't break up the bricks.
