I keep seeing this question asked so.....



Let's find out.

Alright! So, my roommate happened to come home
and agreed to take photos as I did the dang thang!
Hope you enjoy the action shots!

Flaming the utensils.
For this operation I used my scalpel and a pair of long ass tweezers.



Place in first sterile water jar and shake.
Hoping to dilute and remove nasties!
Remove with flamed tweezers.
Place in next jar and shake.
hoping to further dilute and remove nasties.
Remove with flamed tweezers and place on rack.




Flame scalpel and proceed



It wouldn't peel at all.
I guess that's why they call it cracker dry.



It's bound to be a contam bonanza but...
That's not what this is about.
And i'm pretty good with agar so
it'll be easy enough to clean.



For the last plate I tried grabbing with the tweezers
and then hydrating in a fresh sterile water jar.
I thought it'd be interesting to see how it worked.
I only grabbed a tiny piece but... It doesn't take much.



Then I wrapped with glad
as I always do.



That was a mess!



And now we wait

Alright folks, We are back from the oh so nerve racking shroomery hack
and here is the update on the cracker dry cloning attempt.

This is the tissue sample that i soaked in sterile water in an attempt to re hydrate the sample.
No visible growth of any kind. I think. you probably can't tell from the pic but it is possible that we have tiny tiny growth that could possibly be cube myc. I kind of doubt it but time will tell.



Here we have a sample that wasn't hydrated showing nothing.



And another that wasn't hydrated showing plenty of bacterial growth that was carried from the dried fruit via the tweezers.



And another non hydrated sample that exploded with growth!!!



Yeah, Isn't that nice. Maybe i can clean it up.
HA. Noobs may not get the joke but that's pure bullshit.
Nothing but mold.

I'm not done trying this. I really get off on experimenting and learning so I'll try again for sure. My theory so far, Fruits dehydrated at high heat are dead. Any growth is either from spores or contams that landed on the fruit body after dehydration.

I did clone a dry fruit when i was a noob but i can't be certain that the dehydration heat was high enough to kill the myc. That was way back when and i didn't monitor the heat of that dehydrator. I know for a fact that this fruit was dried until it quit loosing weight at a temperature of 165.

It's also quite possible that my noob self got growth from spores and thought i had achieved cloning a dried fruit. Now a days I'm all about careful observation and note taking. It helps me learn. Back in the day though, not so much.

I'll update the op with all helpful as I go. Water is sterilizing and the hood is scrubbing as we speak. My plan is to rinse the fruit in jar after jar of sterile water. Not sure if it'll really help but my hope is to rinse off contams and up my chances of success. I'm still coming up with a game plan but I'll probably rinse, move to new jar, rinse, take a sample or two, and then take a new sample and hydrate it in sterile water before putting to agar. We'll see though. I'm a beer or three deep but.... I'm a professional.


I dehydrate at 165 degrees and don't quit until they quit losing weight.

Maybe before you soak it in the water, try breaking that veil and X-acto a tiny sliver of gill flesh (ideally, this flesh should have a lot less contams due to the unbroken veil protecting it from the elements). Toss that flesh onto agar, i'd bet money Myc grows! Atleast enough to transfer to a new plate, etc.

Either way... awesome that you're doing this! Will be watching with hopeful eyes. 😍

I live somewhere where it's tough to get spores, so tried this a long time ago with a B+ stub. The same water after water bath and everything. My sample was still dirty as hell when I put it on agar, but it ultimately worked well enough. It just took a lot of effort. I'm interested to see how it works out for you.

Sonic's "I'll buy a pressure cooker someday" Instant Pot times and settings:
  • Popping Corn Rapid Re-hydration - Half fill all containers with corn. Empty into Instant Pot. Cover with water. Run 30 minutes on PC. Release, remove, dry.
  • Wheat/Rye Berry Soak Re-hydration - Half fill all containers with grain. Empty in pot with water. Soak for 6-12 hours, air dry until surface is completely dry.
  • Grain Jar Sterilization - Run 2 - 2.5 hrs on PC
  • PastyPlates, MEA Formulation- Run 45 minutes on PC
  • LME Liquid Culture - Run 45 minutes on PC

Spores could possibly germ if you do it this way which would fuck up the test

So how goes it? I expect not so well because of the method of dehydration. The temperature should be enough to kill it but I can't say I've ever actually tried cloning one like that.

I've personally tried reviving dehydrated fruits with no success, but naturally desiccated fruits I've had success on more than one occasion. I've even transferred an entire desiccated plate once, and it took off just fine.
