Cleaning very VERY contaminated jar on agar
    #26119291 - 07/20/19 06:51 AM (4 years, 8 months ago)

Well... i think i have a very unrealistic idea but im willing to give it a go so i can learn something

I have a galindoi jar that is heavily contaminated and has been sitting for about 2 months in a shelf. Recently i picked it up and thought about putting it on agar to see if i can keep it alive.

Heres a pic, what do you think? should i even bother? I am not very experienced with agar. I would buy some spores i just want to learn agar

Thank you all!

looks like atl#7 to me. i dont see contams. What makes you think its heavily contaminated?. You can certainly put some on agar and see if there is bacteria.

Is there metabolites in the jar? can you take different pictures. Do you see any brown veins along the glass?
By 2 months you should have seen stones.

Its was improperly sterilized (steam sterilized actually i know it was insufficient) and has a little pool of water, also there are far too many metabolites and no stones.

Thats why i know it is contam'd.

I would like to know how likely it would be that i could save it. I dont want to have to waste 10+ petris on something truly out of my hands!

Thabk you for your answer mate

It looks like the right color for atl. If you can find a stone its a great candidate for a iso. and you should be able to get clean growth with out to much work. When you open the jar let us know how it smells. Have you worked with atl before?

Sory man, i missed that part of your anwswers, here are some better pics.

I would give it a try on agar then. From where should i take my sample? There would appear to be some clusters of mycelium, like attemps to make sclerotia but nothing significant.
We could say its my first time working with it seriously

If it  was me i would take some wispy aerial mycelium off the lid or jar and make a plate or two. I would also break up the spawn and find a stone and place it on. I would also try and find a larger stone that i could break open and potentially get a inner stone sample.
The spawn could still fruit. 1 more month would also make any stones in there a bit bigger.
The myc should have a nutty smell.
