Emergency Noob Cloning Question

Hi!

I am new to this practice and have been working with some PF Tek grows since early this year. Back in early June I inoculated 12 BRF cakes with 1/2 Golden Teacher and 1/2 Cambodian spores. The jars all colonized well, but then we had some really hot, humid weather, and they all got damp through their dry verm barriers. Out of those jars only 2 of each strain survived uncontaminated.

My cambodians are finally fruiting and today I realized that one of the cakes appears to be an albino strain. I'm taking a spore print to make sure it isn't just a weird infertile mutant strain, but I'm pretty sure it's an albino.

The fruits aren't very big but the pinsets are good, and I'd like to propagate the strain if I can as I know they're a rarity. However, in my last 4 experiments making sterile spore syringes I've had a staggering 0% success rate, so I figure that I need to learn how to clone these things, and fast, if I want to preserve them.

I have a SAB that I made for my spore print experiments. The arm holes are kind of big and they don't have any kind of tubing affixed to them to limit airflow. It's just 2 holes I carved into a sterilite bin with a dremel :shrug:  I wipe the inside down with alcohol before I use it and pour a little soapy water in the bottom, but that's as far as I go with sanitizing it. I do wear gloves that I've sprayed down with alcohol while I'm using it, but I'm wondering if this approach or design isn't flawed since I'm having a lot of problems with contams, even with BRF cakes I inoculated inside of it (which actually wasn't ever an issue when I was 'noccing in open air...) so any tips there would be appreciated.

Anyway; I've never worked with agar and don't really have the funds for it right now. Since I wanted to get started with cloning experiments right away, I used cloneufc's Easy Clone Tek; I wasn't sure how big to make my biopsies so each one is about 1 1/2" long by 1/2mm thick. I didn't have much source material to work with as so far only a few of the cambodians were big enough to pick. I have maybe 8 samples in plastic baggies in a cool, dark drawer now (relatively cool that is - its about 90 degrees here on average right now).

My first question is - what should I do if they actually work and start growing mycelium? I'm not really sure what kind of grain jars to transfer to, or how to direct transfer to grain (I've only ever used spore syringes before). Do I just sprinkle the cultures into fresh BRF cakes inside my SAB? Should I try rye or birdseed or something else instead? Do I shake it up once I drop the myc in there, or let it sit on top? How do I keep this from getting contaminated?

My second question is - is this the best way to be going about cloning? Is there a better clone tek for noobs that I'm not aware of with higher success rates?

My last question is, if I did manage to get a sterile spore syringe going, would the resulting cakes be albino as well? I've heard that spores can be kind of a crapshoot when it comes to genetics. And if that would work, could someone please give me some tips on better techniques for sterile spore syringes? I always boil my water for 10 mins first and then let them cool to room temp. I put my mushroom caps directly into sealed glass jars I've cleaned with 70% ISP and let them sit out overnight, then get into my SAB and mix the sterile water with the print inside the jar... for whatever reason the resulting syringes always fail to produce mycelium in any of the jars.

Any help is sincerely appreciated.

2. I know it's not what you're looking for, but the best cloning Tek for noobs involves agar.

3. Spores are for sexual reproduction which involves varying genetics, so you can indeed wind up with non-albino strains from spores taken from an albino strain.  It's still possible to get albinos as well, and further inbreeding them will result in very low chances of not getting albinos, but that itself will introduce other issues. Genetic diversity is generally good.
