Personally I've never done agar up until now, finally perfected my plates! (used to be 10/10 bacterial plates, now its 0/10 :lol:)

I've always been a bit cheeky and just MS inoculated everything, 12/12 of my rye quart jars look beautiful, little bit of yellow excretion but no contam signs. Never had a contam personally from MS inoc however may just be beginners luck :lol:

However I'm sick of MS genetics and want to dive into agar to get some proper pinsets and canopies.

OK. Thanks. Good to know. It's just surprising to me, in that event, that not one of my jars developed ANY mycelium whatsoever. Now it's a fact that the spores came from Arizona, and could have been sitting in a hot warehouse, and when they arrived, were not immediately refrigerated. That could have been the main factor. I don't know. I just doubt that this would have killed all the spores, but maybe anything is possible.

Good question. Actually, not all the jars showed contamination. A few showed contams within the first two weeks. Then more in the third week. But some didn't show green mold, but nevertheless, not even a sign of any mycelium whatsoever. This leads me to believe that the spores were duds.

I've never done agar before, but in all the process videos and instructions I've read, trying to get a clue, there seems to be one crucial point that I profess ignorance of, that infuriatingly, nobody seems to home in on, or explain. And that is, are we, or are we not, supposed to inoculate precisely before the agar solidifies?

I saw one video where the guy has all the expensive equipment, real scientist in appearance, and obviously flush with money and all, and as soon as the agar has cooled, he does fuck all with it except wraps the petri dishes in parafilm, smiles, acts like we're done here, and puts them for a rainy day. Huh?? WTF?

I mean once the agar has cooled, how do you leverage it? I mean, if you scrape spores onto the agar, they're just going to lie on a dry surface, right? How is that going to integrate with the agar or get it to interact with your spores? How is that going to do anything to develop mycelium?

Now if you use a spore syringe, again, how does that work? It's just a drop of spore water, yeah, that's lying on the surface of the agar, where presumably the solidified agar has a near impenetrable surface. So again, unless you apply the spores or spore water just before the agar solidifies, how is this supposed to get mycelium going? It doesn't appear likely to me. Plus, a drop of water from a syringe lying on impenetrable agar for 8 days seems like a sure recipe for contamination to me. So can somebody kindly tell me how this is really supposed to work? I don't want another pointless failure.

agar media contains a lot of moisture - agar media is mostly water, like Jello

Aerobic organisms don't need to penetrate into the agar, they are fine sitting on top of it and eating the surface. That's why any little living contaminant floating in the air that lands on your agar is going to start growing

but hang on, aren't you supposed to drag an inoculation loop zig zagging over the surface. I thought the purpose of that was to sort of embed the spores over the surface, to embed them within the agar.

The point is to thin out the spores so there are multiple colonies to transfer from as opposed to one big fluff ball. Better chance at transferring away from contams in fewer tries this way.
