Are these plates good for A2G?

I’m new to working with agar and would like some input on whether these plates look good to transfer to grain. They’re both second transfers from tissue scraped from the inside of a young Burma variety.

What about 6:00 on the left plate, is that disorganized growth a concern?

Looks fine to me. Granted the jars he knocks up might not want to be masters. It might not be a mono culture but he could do some grain jars with the plates.

Might not want to transfer that particular part to grains.  But I hear there's no real way to tell what a strain(s) will do until you grow it out.  With that said, the wispiness makes it look weak by comparison don't you think?

Now that I look at it again the growth between 8:00 and 10:00 on that left pic seems to scatter and get patchy towards the outside. Feels shady to me, I'd take another transfer from a cleaner organized section. I know sometimes you'll see rhizo and tomentose growth on the same plate but I thought it will still have more organization than that if it's clean. But I don't know much and have been riddled with problems lately, becoming suspicious of everything hehe.

I do, but the wrap was messing up the detail around the edges, which is the part I was most curious about. Wrapped it back up right after the photo :smile:

I was also concerned about those wispy parts... would it be reasonable to cut wedges around that part for a transfer to grain (for just one bin)? I’m on a bit of a time constraint. I’ll transfer the best parts of one of the plates to more agar to isolate, for sure.

If you're in a hurry go for it, cut out the clean parts and be sure to not cut very close at all to the shady looking parts or the center as it may have grown out from the center into those areas

Does anyone have a link to the source of that belief?  How solid is that?  Im sure nice clean cultures have organized growth but why doesn't what we see on your plate just indicate multiple strains working through each other?

I don't see any contamination, like a liquid or discolouration or something.

around 9:00 toward the edge on the left plate doesn't look like interwoven culture it looks kind of like interrupted culture to me. I may be wrong and just projecting my fears and traumas onto his dish :rofl:

These are multiple strains showing tomentose and rhizo growth, but to me they are still organized
