Newbie question
    #26093209 -

Hi everyone,

I hope this finds you all having as fantastic a day as you could hope to have, whatever your circumstances.

This is my very first post on shroomery (long-time lurker). My friend, however, needs advice on his first mono tub grow. Her 66 ltr monotub bulk-substrate of a manure/coir/verm mix was inoculated with 4lbs of ryegrain spawn on June 26th and was a bit slow to colonize.

It has been incubating at 78F and 99% humidity since that time and whilst the mycelia seem healthy and strong, there are pools of yellow goo on top and at the sides of the substrate. She would like to know if these are metabolites or a likely bacterial contamination.

Your thoughts and experience will be invaluable.

Metabolites are often the result of bacterial contamination.

"I thought to myself 'Boy, I'm sure glad there's nobody here to see this because this is exactly the sort of thing that gets people riled-up and they assume you're dying and that something has to be done. Where if you're alone, you know, you either come through it or you die, but in any case you avoid the fuss.'"

Hopefully pics should show now:

I would add that there is no sweet, sour or bitter odour coming from the tub either.

If the metabolites are released in response to bacterial contamination, should she abort this entirely?

Soak up the yellow and let it keep growing. I've gotten some nice fruits from tubs looking like that.

That looks pretty bad, but unless you got five more going there's no point in tossing it now. You can still get a flush or two, even if it's bacterial.

I'd advise you to stop misting entirely for now, keep it a bit on the dry side to give the mycelium an edge. Maybe move it somewhere cooler, if you can.

Thanks for the advice Crackatoa - I’ll let her how and see what happens from here!

Yeah that’s what I thought when I saw it too ColdTurkey... It incredibly disheartening as I know how much effort she had made to ensure sterility throughout each of the stages.

I’ll get her to do what you and Crackatoa have each said; soak up the excess metabolite, stop misting and remove lower the incubation temperature.

Hey all,

Long time no speak! Thanks for those who replied to me last year. It really helped and I’m pleased to say that all I needed was patience and time. I got some very healthy fruits and 4 flushes before contamination began to inhibit anything further.

(I’ve posted pics to show my results).

I’m now looking to start my second grow and would like some opinions on my agar plates. Ideally I would have posted this as a new topic, but I don’t have the permissions haha.

That would help wouldn’t it!

I’m keen to know how they  are looking and which look most promising. Of interest is one spot on plate 3 that has a miniature tree - branches and all - growing out of the agar and is touching the upper plate.

Let me know your thoughts ?

Im not sure if you can see, but i marked in blue where I would start transferring from

is this a germination plate? Spore syringe to agar?

AGAR ENVY! (Anything and All things agar!)

you should post your plates in the linked thread as well, its a pretty active thread where you can better feedback then I provided on your plates

Thanks so much and I appreciate the feedback! They are indeed germination plates. I’m assuming the next steps will be to begin to isolate on new, sterile plates?

Unfortunately, it says I’m not allowed to post a new topic, so I can’t post in the section. I really appreciate you taking the time to share the knowledge you have man.

its not making a new topic, its just "commenting" on that thread.

you should be able to post there.

yes the next step would be to transfer from those plates to new ones and allow it to grow out.

c10's Agar Guide + Tips & Tricks for reference to what your looking for when making transfers and assessing whether or not the culture is clean

no problem man, always willing to help if I can

what variety are you working with?

I see, sorry - I’m new to being an active member on a forum. I usually just lurk and read, but with a hobby like this interaction is very important (or so I am finding).

I’ll check that out and again, thanks for the really good steer man.

In terms of variety, I grew McKennaii cubes last year and they were fun. My liquid culture that I bought died though so this time I’m growing PES Amazonian. Do you have experience with either?
