90c/kg organic rye . . . help them out.

I have 2x 500ml 1x 250ml 1x 100ml media bottles, mainly because I started with a small PC. Helpful if you are doing different agar for spores, clones and transfers.

As Inski says, pour when cool enough (or pre pour glass petri). Many in here will say microwave agar bottles, I don't like microwave.

Note, plastic petri are one use only (i.e. you can't re-sterilse petri unless you have a gamma radiation source). Pour the entire sleeve in one go to avoid contamination.

Damn, That's some cheap grain. I'll need to keep that in mind! Hopefully can grab some of that soonish, although what's shipping like?

UV should be plenty fine to sterile petri dishes :P One of the flow-hoods in my old lab had a UV sterilisation setting that worked for things like plastic petri dishes. Never really got used tho.

If you have a flow hood, you can open the sleeve in it and then tape it shut still in the flow hood and you won't get contams. IDK about a SAB tho.

I assume also that if rye is unavailable at a decent price, wheat/oats/barley is still acceptable? Looking around it seems that people use rye because it's cheap and easy to access? EDIT: 2 seconds after posting this I find:

They are in canterbury... Find someone passing. . . Though 25kg was $43 deliverred to auckland. Yes folks thats organic rye deliverred for under $1.70/kg. Enough to keep me happy for a year or so . . .
@inski, what contaners do you use to spawn and grow. It seems that you use smalk round containers for your grows. Do you make small spawn jars, or many substrate jars from a quart of spawn?

Depends entirely on the amount of work you're doing. If you have 40 plates, 1000mL agar, and your tools to autoclave, then it would probably be needed. If you're just doing 6 plates, 150mL agar and your tools then it's wayyyy over the top haha.

And stovetop isn't too bad. Get it to pressure, turn the temp down, set a timer, wander off, come back when the timer goes off and turn the stove off.

Mitre10 sell WBS (fortunately missing corn kernels)

Interlab sold 75mm glass petri for $3 each if you ask for their best price, maybe better?

Media bottles, the more expensive ones (locally) have better seals and can handle a PC run while sealed if cooling naturally. If buying from China, you are rolling the dice. Please report back if you get a good roll.

I found a 36L Pressure cooker in NZ for $215 (Click Here).


Working pressure 50 KPa? *math* ah so this is one of those 7KPI models.
From my understanding it works but you have to run it for twice as long?

You don't need expensive bottles, the seal doesn't matter because you always leave the lid loose when sterilising agar, just cover the top with foil.

Yes, this cooker runs at roughly 7.252psi which is about half the recommended pressure for sterilisation so you would have to run it twice as long.

As long as you leave your media bottle somewhere clean.  When they cool the air inside cools, sucking surrounding air into the bottle. I open my PC when the saftey drops, and screw the lid closed, and transfer to the SAB.

Yes, you might run into problems working in those conditions.
I open my pc in front of the laminar flow hood so I don't worry about tightening the lid.

What about for grain work and LC?

I was thinking that once I've got some nice myc, I'd get some LC going and inoculate grains with it. I get that I can just pour it, but my understanding is that there's a way higher chance of contam doing that?

You can check the internet for needle exchange pharmacies, who will happily exchange needles and syringes. These are tiny and inappropriate for microscopy. The whole idea of the needle exchange is to maintain anonymity so IV users aren't scared away and stay safe. You can get 10ml syringes, though most are luer slip (great for dosing the kids with paracetamol), not luer lock, I prefer the locks for security. You may have trouble finding 16 gauge needles also, which are the fat ones required when working with culture filled fluids.

Look at bodhisata and pastywhyte for their LC methods. Pouring works fine.

If you do need needles, capes medical supplies are about 10% of the cost of pharmacies but you have to buy 100. So, buy 10 get 90 free. Pharmacy 10ml syringe $2.30. capes, 100 x syringes with 100 x 16g 40mm needles for <$33.
